含红芪与含黄芪的玉屏风散对SAMP8小鼠脾T细胞及PI3K表达的影响 被引量:5

Influence of Yupingfeng Powder containing Radix Hedysari or Radix Astragali on splenic T lymphocyte and PI3K expression in SAMP8 mice

摘要 目的:比较含红芪与含黄芪的玉屏风散对SAMP8小鼠脾T细胞及PI3K表达影响。方法:取40只SAMP8小鼠随机分成4组:玉屏风散红芪组、玉屏风散黄芪组、胸腺肽组、SAMP8组,每组10只,另取10只昆明种小鼠作为青龄鼠组,其中SAMP8组、青龄鼠组予以相同体积的0.9%氯化钠溶液代替灌胃。连续给药14d后,测定各组血清细胞因子、脾T淋巴细胞表面分子、PI3K mRNA及其蛋白表达水平。结果:与SAMP8组小鼠比较,各给药组小鼠血清IL-2、IFN-γ含量均明显增高(P<0.05),且玉屏风散红芪组IL-2含量高于玉屏风散黄芪组(P<0.05);中药组CD8分子表达增高,胸腺肽对于CD8分子表达无影响。各给药组CD4分子、CD28CD152双阳性表达均下降(P<0.05);各给药组PI3K mRNA表达量均上调(P<0.05),玉屏风散红芪组与胸腺肽组效果相当且优于玉屏风散黄芪组(P<0.05);各给药组PI3K蛋白表达量均上调(P<0.05),但中药组不及胸腺肽组(P<0.05)。结论:含红芪与含黄芪的玉屏风散黄对SAMP8小鼠均具有一定的延缓免疫衰老作用,且含红芪的玉屏风散在提高SAMP8小鼠IL-2含量和上调PI3K mRNA表达方面优于含黄芪的玉屏风散。 Objective: To compare the influence of Yupingfeng Powder containing Radix Hedysari or Radix Astragali on splenic T lymphocyte and PI3 K expression in SAMP8 mice. Methods: Forty SPF grade SAMP8 mice were randomly divided into four groups(n=8) including Yupingfeng Powder containing Radix Hedysari group, Yupingfeng Powder containing Radix Astragali group, Thymosin group and SAMP8 group, and ten Kunming mice of clean grade were token as young group, among which both the SAMP8 and young groups were gavaged with 0.9% sodium chloride solution of the same volum. After fourteen days of drug administration, measure the level of serum cytokines, the expression of splenic T lymphocyte surface molecules, the PI3 K mRNA and the PI3 K. Results: Compared with SAMP8 mice with no treatment, the serum cytokines IL-2 and IFN-γsignificantly rised in SAMP8 mice with treatment, especially that the IL-2 of Hedysari group was more significant than the Astragali(P<0.05). The expression of surface molecule CD8 rised in TCM groups, whereas there was no effect in the Thymosin group. The expression of surface molecule CD4 and CD28CD152 both decreased in treated groups; the expression of PI3 K mRNA rised in treated groups, especially that the Hedysari group was more significant than the Astragali(P<0.05); the expression of PI3 K rised in treated groups. Conclusion: The Yupinfeng Powder containing Radix Hedysari or Radix Astragali both has the function of delaying immunosenescence, and the effect of Yupinfeng Powder containing Radix Hedysari is more significant than containing Astragali in raising serum cytokine IL-2 and the expression of PI3 K mRNA.
