佐木阿汤对缺氧大鼠心肌细胞Ca^(2+)浓度及Cx43表达的影响 被引量:5

Effects of Zuomua Decoction on concentration of Ca^(2+) and expression of Cx43 in myocardial cells of hypoxic rat

摘要 目的:探究佐木阿汤对缺氧大鼠H9C2心肌细胞Ca^(2+)及缝隙连接蛋白43(Cx43)表达的影响与机制。方法:将H9C2心肌细胞株分为正常组,模型组,5%、10%、15%佐木阿汤组,依那普利组和生理盐水组。通过含药血清干预与建立缺氧模型,应用CCK-8比色法检测细胞存活率,荧光酶标仪法检测心肌细胞内Ca2+浓度变化,Western Blot法测定心肌细胞中Cx43与p-Cx43表达。结果:(1)细胞存活率:15%佐木阿汤组、依那普利组显著高于模型组(P<0.05);(2)Ca^(2+)浓度:5%、10%、15%佐木阿汤组、依那普利组显著低于模型组(P<0.05);(3)Cx43与p-Cx43:15%佐木阿汤组、依那普利组表达显著高于模型组(P<0.01)。结论:佐木阿汤可能通过降低细胞内Ca2+浓度,提高Cx43与p-Cx43表达,改善细胞间隙通讯,对缺氧心肌细胞具有一定的预防和保护作用。 Objective:To explore the effects and mechanism of Zuomua Decoction on concentration of Ca^(2+) and expression of Cx43 in myocardial cells of hypoxic rat.Methods:H9C2 myocardial cellstrain were divided into normal group,model group,5%,10%,15% Zuomua Decoction groups,10% enalapril group and physiological saline group.The hypoxic model was established by the intervention of serum containing drugs,cell viability and survival rate was determined by CCK-8 colorimatric method.The changes of Ca^(2+) concentration in myocardial cells were detected by fluorescence labeling method.The expression levels of Cx43 and p-Cx43 were determined by the method of Western Blot.Results:(1)Cell survival rate:Compared with the model group,15% Zuomua Decoction group and enalapril group were obviously increased(P<0.05);(2)Ca^(2+) concentration:Compared with the model group,5%,10%,15% Zuomua Decoction group and enalapril group were decreased significantly(P<0.05);(3)Cx43 and p-Cx43:Compared with the model group,15% Zuomu Decoction group and enalapril group were significantly increased(P<0.01).Conclusion:Zuomua Decoction has significant cardiomyocytes protective effect against hypoxia injury.It may increase the expression of Cx43 and p-Cx43 and decrease the intracellular Ca^(2+) concentration in hypoxia myocardial cells,which can improve the intercellular communication.
