摘要
目的:研究知柏地黄汤对解脲脲原体(UU)感染模型大鼠精子线粒体Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶的影响。方法:80只雄性SD大鼠,随机分成假手术组、模型组、中药组(知柏地黄汤8.56g生药·kg-1·d-1)、西药组(强力霉素20mg·kg-1·d-1)、中西组(知柏地黄汤+强力霉素),除假手术组外,其他组均采用经大鼠膀胱注射UU建立UU感染动物模型,连续灌胃21d后处死,取标本检测精液子质量,分光光度法检测精子线粒体Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活性。结果:与假手术组比较,UU感染模型大鼠精子浓度、精子活动率,a+b级精子百分率及精子线粒体Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活性下降(P<0.01);治疗各组中精子活动率,a+b级精子百分率及精子线粒体Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活性上升。结论:知柏地黄汤能够提高UU感染大鼠精子线粒体Na^+-K^+-ATP酶、Ca^(2+)-Mg^(2+)-ATP酶活性。这可能是知柏地黄汤治疗UU感染所致不育症的机制之一。
Objective:To research the effects of Zhibai Dihuang Decoction on activity of Na^+-K^+-ATP enzyme and Ca^(2+)-Mg^(2+)-ATP enzyme in spermaticmitochondria of ureaplasma urealyticum(UU) infected rat model.Methods:Eighty male SD rats were randomly divided into 5 groups:sham operation group,model group,Chinese medicine group(Zhibai Dihuang Decoction 8.56 g crude drug·kg-1·d-1),western medicine group(doxycycline 20mg·kg-1·d-1),integration of traditional Chinese and western medicine group(Zhibai Dihuang Decoction + equivalent dose of doxycycline) respectively.Except for the sham operation group,ureaplasma urealyticum infected rats were established in other groups by bladder injecting ureaplasma urealyticum.The corresponding medicine were separately administrated by gavage for 21 days.The sperm quality was detected,and the activity of Na^+-K^+-ATP enzyme and Ca^(2+)-Mg^(2+)-ATP enzyme were detected by spectrophotometry.Results:Sperm concentration,sperm activity rate,grade-a and-b motility and mitochondrial Na^+-K^+-ATP enzyme and Ca^(2+)-Mg^(2+)-ATP enzyme activity decreased in UU infected model group,which were significantly lower than the the sham operation group(P<0.05);whereas those were significantly reseved by the treatment of Chinese medicine,western medicine and integration of traditional Chinese and western medicine.Conclusion:Zhibai Dihuang Decoction can improve spermatic mitochondria Na^+-K^+-ATP enzyme and Ca^(2+)-Mg^(2+)-ATP enzyme activity in UU infected rats,which could be the underlying mechanism of Zhibai Dihuang Decoction treating infertility induced by UU infection.
