温肺降浊方对血管性痴呆大鼠海马神经元凋亡的影响

Effects of Wenfei Jiangzhuo Formula on hippocampal neurons apoptosis in vascular dementia rats

目的:探讨温肺降浊方对血管性痴呆(VD)大鼠海马Cytc、Bax蛋白表达的影响及其神经保护作用。方法:SD大鼠120只,随机分为假手术组、模型组、温肺降浊方组(中药组)、盐酸多奈哌齐组(西药组),每组30只。采用2-VO法制作VD大鼠模型,Morris水迷宫测试大鼠学习、记忆能力,流式细胞术检测海马细胞的凋亡情况,免疫组化法检测Cytc、Bax蛋白的含量。结果:(1)与假手术组比较,模型组大鼠学习、记忆成绩下降;与模型组比较,中药组与西药组大鼠的学习与记忆成绩均有显著提高(P<0.05);(2)与假手术组比较,模型组细胞凋亡发生率显著增加(P<0.05);与模型组比较,中药组及西药组细胞凋亡率均显著下降(P<0.05);(3)与模型组比较,中药组与西药组Cytc、Bax蛋白表达均显著下降(P<0.05),且中药组与西药组比较无统计学意义。结论:温肺降浊方能明显改善VD大鼠学习记忆能力,其作用机制可能与其通过抑制Cytc、Bax蛋白的表达,从而抑制海马神经元的凋亡有关。

Objective:To observe the effects of Wenfei Jiangzhuo Formula on the expression of cytochrome C(cytc) and Bax proteins in vascular dementia(VD) rats and its neuroprotection.Methods:A total of 120 male SD rats were randomly divided into four groups:sham-operation group,model group,Wenfei Jiangzhuo Formula group,and donepezil group.Transient global cerebral ischemia model was made by 2-VO technique.The memory and learning behaviors were measured by morris water maze.The expression of Cytc and Bax proteins were detected by immunohistochemical method and the amount of apoptosis cells was measured by flow cytometry.Results:The memory and learning behaviors were decreased in the model group when compared with the sham group,which were improved by the treatment of Wenfei Jiangzhuo Formula and donepezil(P<0.05).The results of flow cytometry showed that the transient global cerebral ischemia significantly increased the ratio of neuronal apoptosis,but it was reversed by the treatment of Wenfei Jiangzhuo Formula and donepezil(P<0.05).The results of immunohistochemistry showed that the expression of Cytc and bax proteins were decreased in the Wenfei Jiangzhuo Formula group and donepezil group(P<0.05),but there was no statistical difference between the two groups.Conclusion:Wenfei Jiangzhuo Formula could significantly improve the memory and learning behaviors of vascular dementia rats,which might relate to inhibit the hippocampal neurons apoptosis through decreasing the expression of Cytc and Bax proteins.
