摘要
目的:筛选桂附地黄丸抗中老年男性部分雄激素缺乏综合证(PADAM)的有效部位,并对有效部位的成分来源进行归属分析,为深入研究桂附地黄丸治疗PADAM的药效物质提供基础。方法:(1)制备桂附地黄丸的石油醚、氯仿、乙酸乙酯、正丁醇、水提部位,采用腹腔注射环磷酰胺致大鼠PADAM模型,以大鼠体质量、血清总睾酮(TT)、游离睾酮(FT)、肾脏SOD、MDA为主要评价指标,筛选桂附地黄丸抗PADAM的有效部位;(2)建立有效部位的指纹图谱,并与复方单味药及相应缺味药比较,分析有效部位成分的来源及归属。结果:(1)与正常组比较,模型组大鼠体质量、血清TT、FT含量和肾脏SOD活性显著降低(P<0.01),肾脏MDA水平明显升高(P<0.01)。桂附地黄丸、水提、水+正丁醇、水+乙酸乙酯、水+氯仿、水+石油醚部位能不同程度地改善PADAM大鼠的症状(P<0.01,P<0.05),其中水+正丁醇部位的作用更加显著(P<0.01)。(2)建立了水+正丁醇部位的指纹图谱,确定了以马钱苷为参照物的16个共有峰。(3)初步判断了桂附地黄丸水+正丁醇部位的16个特征峰的归属,此部分化学成分主要来源于熟地黄、牡丹皮、泽泻和山茱萸,以山茱萸所含的特征峰最为丰富。结论:水+正丁醇部位作为桂附地黄丸抗PADAM的有效部位,药效明确,质量可控,实验结果可为进一步研究其药效物质提供实验依据。
Objective:To Screen the effective parts and analyze the component ascription of anti androgen deficiency syndrome in the aging male(PADAM) in Guifu Dihuang Pill,in order to provide the basis for the further study on the effective substance of Guifu Dihuang Pill for treating PADAM.Methods:(1)The petroleum ether,chloroform,ethyl acetate,n-butanol and water extractions of Guifu Dihuang Pill were prepared,and the PADAM rat models were established by adopting intraperitoneal injection of cyclophosphamide.The effective parts fro anti PADAM in Guifu Dihuang Pill were screened based on the indexes as body weight,serum total testosterone(TT),free testosterone(FT),renal SOD and MDA.(2)To establish the finger-print of effective parts,and analyze the component attribution of effective parts based on comparison of single herb in compound prescriptions and corresponding void herb.Results:(1)Compared with control group,the body weight,serum levels of TT,FT,and SOD like activity in kidney of rats in model group decreased significantly(P<0.01),and MDA level in kidney increased significantly(P<0.01).The parts in Guifu Dihuang Pill extracted by water,water+n-butyl alcohol,water+ethyl acetate,water+chloroform,water+petroleum ether were all could improve the symptoms of PADAM rats in different extent(P<0.01,P<0.05),of which the effect of water+ n-butanol alcohol was the most obvious(P<0.01).(2)The finger-print of parts extracted by water+n-butanol alcohol was established,16 peaks were confirmed with the comparison of loganin.(3)The analysis on the affiliation of 16 characteristic peaks of parts of Guifu Dihuang Pill extracted by water+n-butanol alcohol showed that the chemical components of these parts mainly came from prepared rehmannia root,tree peony root bark,oriental waterplantain rhizome,and asiatic cornelian cherry fruit,of which the characteristic peaks contained in asiatic cornelian cherry fruit was the most abundant.Conclusion:The parts in Guifu Dihuang Pill extracted by water+n-butanol alcohol were the effective parts for anti-PADAM with clear effects and controlled quality,and the experimental results could provide experimental bases for the further study on its pharmacodynamic material.
