龟板联合阿伦磷酸钠对激素性骨质疏松大鼠腰椎Runx2、CTSK表达的影响 被引量:14

Effects of tortoise plastron in combination with alendronate on expression of Runx2 and CTSK in lumbar vertebrae of rats with glucocorticoid-induced osteoporosis

摘要 目的:观察龟板联合阿伦磷酸钠(ALN)对激素性骨质疏松(GIOP)大鼠腰椎Runx2、CTSK表达的影响。方法:40只3月龄雌性SD大鼠随机分为4组:空白组、模型组、ALN组、龟板联合ALN组(联合组)。模型组、ALN组及联合组皮下注射地塞米松造模,成功后分别用0.9%氯化钠溶液、ALN和龟板联合ALN进行灌胃。12周后取大鼠腰椎进行:micro-CT检测腰椎骨微细结构、HE染色观察骨组织形态学,qPCR及Western blot分别检测腰椎Runx2、CTSK mRNA和蛋白表达。结果:与空白组比较,模型组Tb.N、Tb.Th明显下降(P<0.01),Tb.Sp、CTSK mRNA和蛋白表达明显升高(P<0.01),Runx2 mRNA和蛋白表达呈下调趋势;与模型组比较,ALN组和联合组BS/TV、Tb.N、Tb.Th、Runx2蛋白表达明显升高(P<0.01),Tb.Sp、CTSK mRNA和蛋白表达明显下调(P<0.01)。结论:龟板联合阿伦磷酸钠抗大鼠腰椎GIOP的协同机制可能与调节Runx2、CTSK的表达密切相关。 Objective: To study the effects of tortoise plastron in combination with alendronate(ALN) on expression of Runx2 and CTSK in lumbar vertebrae of rats with glucocorticoid-induced osteoporosis. Methods: Forty 3-month female SD rats were randomly divided into four groups: control group, model group, ALN group and tortoise plastron in combination with ALN group(combined group). Model group, ALN group and combined group were received hypodermicinjection of dexamethasone to establish model, and then were treated with 0.9% sodium chloride solution, ALN and tortoise plastron in combination with ALN by gavage for each group. Lumbar were collected for analysis after 12 weeks. Micro structure of lumbar was detected by microCT. Morphological tissue of lumbar was observed by HE staining method; Runx2, CTSK mRNA and protein expression were detected by qPCR and Western blot. Results: Compared with control group, model group showed significantly decreased Tb.N and Tb.Th(P<0.01), significant increased Tb.Sp, CTSK mRNA and protein expression(P<0.01), and decreased trend of mRNA and protein expression of Runx2. Compared with model group, ALN group and combined group showed significant increased BS/TV, Tb.N, Tb.Th and protein expression of Runx2(P<0.01), and significant decreased Tb.Sp, CTSK mRNA and protein expression(P<0.01). Conclusion: The cooperationmechanism of tortoise plastron in combination with ALN on treating lumbar GIOP of rats might be closely associated withthe expression of Runx2 and CTSK.
