摘要
目的:研究化浊解毒活血通络方对脑缺血再灌注损伤模型大鼠神经细胞凋亡相关基因Bcl-2及Bax表达的影响,探讨其神经保护作用机制。方法:采用改良的Zea-Longa线栓法通过阻断大鼠右侧大脑中动脉制造局部梗死性脑缺血再灌注损伤模型。运用TUNEL法标记凋亡细胞统计阳性细胞数并计算凋亡率,应用RT-PCR法检测各组大鼠缺血侧脑组织中Bcl-2及Bax mRNA的表达水平。结果:与假手术组比较,模型组及各用药组细胞凋亡率、Bax表达均明显上升(P<0.05),Bcl-2明显降低(P<0.05);与模型组比较,中药各组及阿司匹林组细胞凋亡率下降(P<0.05),Bcl-2基因表达升高,Bax基因表达降低(P<0.05);在调高Bcl-2基因表达方面,中药3组效果不如阿司匹林组(P<0.05),中药3组疗效与浓度呈正相关;在下调Bax基因表达方面,阿司匹林组及中药中、高剂量组3组表达无显著性差异,中药低剂量组Bax基因表达较其余治疗3组明显较高(P<0.05)。结论:化浊解毒活血通络法可以通过上调细胞凋亡基因Bcl-2、下调Bax表达从而降低细胞凋亡率,发挥其脑保护作用。
Objective: To investigate the effects of Huazhuo Jiedu Huoxue Tongluo Prescription on apoptosis and expression of Bcl-2 and Bax in rats of cerebral ischemia-reperfusion injury, and to explore the neuroprotective effect of Huazhuo Jiedu Huoxue Tongluo Prescription. Methods: The mice model of cerebral ischemia reperfusion injury(CIRI) was established by using Zea-Longa method. The number of apoptotic cells was marked and the neuron apoptosis rate was detected by TUNEL. The expressions of Bcl-2 and Bax in ischemic brain tissue were detected by the RT-PCR method. Results: Compared with the sham group, model group and the treatment group contained significantly high rate of apoptosis and expressions of Bax(P<0.05), the expression of Bcl-2 decreased(P<0.05). Compared with model group, the rate in Aspirin group and each group apoptosis was lower while the expression of Bcl-2 gene was higher, the expression of Bax was lower(P<0.05). In increasing Bcl-2 gene expression, the three traditional Chinese medicine groups were lower than Aspirin group(P<0.05), and the curative effect was positively correlated with the concentration in the three Chinese medicine groups. In decreasing Bax gene expression, the Aspirin group, traditional Chinese medicine medium dose group and high dose group showed no significant difference, while in Chinese low-dose group, the Bax gene expression was significantly higher compared with the other three groups(P<0.05). Conclusion: Huazhuo Jiedu Huoxue Tongluo Prescription can increase the expression of Bcl-2, and reduce the expression of Bax to decrease the apoptosis rate, which might be the mechanism of neuroprotections.
