摘要
目的:研究甘麦大枣汤对"怒"模型大鼠行为学特征的影响。方法:将40只Wistar雄性大鼠随机分成正常组、模型组、甘麦大枣汤组和香芍颗粒组,每组10只。除正常组外,其余各组以社会隔离与居住入侵方法进行"怒"模型大鼠复制。28d完成造模后,各组开始给予相应的干预措施,其中,模型组、甘麦大枣汤组和香芍颗粒组继续以造模方法给予相应的刺激。香芍颗粒组大鼠灌服香芍颗粒药液(浓度为0.1g/m L);甘麦大枣汤组灌服甘麦大枣汤浓缩药液2m L;正常组和模型组灌服等体积的蒸馏水,均1次/d。各组均干预28d。观察体质量变化情况,同时运用旷场实验、攻击行为测试观察大鼠的行为学变化情况。结果:造模28d后,模型组、甘麦大枣汤组、香芍颗粒组较正常组体质量减轻(P<0.01);水平运动距离明显延长(P<0.05,P<0.01);直立次数明显增多(P<0.05);攻击次数、攻击时间显著增多(P<0.01);攻击潜伏期时间明显缩短(P<0.01);攻击行为总分增高(P<0.05)。干预后,甘麦大枣汤组、香芍颗粒组与模型组比较,各项指标改善的差异性显著(P<0.01),而甘麦大枣汤组与香芍颗粒组比较,各项指标改善的差异无统计学意义。结论:甘麦大枣汤通过改善"怒"模型大鼠的行为学特征,对异常"怒"发挥调整作用。
Objective: To investigate the effect of Ganmai Dazao Decoction on behavior characteristics in rats model of ‘anger'. Methods: Forty Wistar male rats were randomly divided into the control group, model group, Ganmai Dazao Decoction group and Xiangshao Granule group, 10 per each group. Expect for the control group, the rat model of anger was induced by social isolation and residential invasion. After 28-day model establishment, the three groups were continuously subjected to the social isolation and residential invasion coupled with the corresponding treatments. The Xiangshao Granule group was given the Xiangshao Granule liquid(concentration of 0.1g/m L) by gavage, once a day. The Ganmai Dazao Decoction group was given the 2m L of concentrated Ganmai Dazao Decoction by gavage. Both of the control group and the model group were given the same volume of distilled water by gavage. Thetreatment process lasted for 28 days. Body weight and behavior tests including open field test and aggressive behavior testing were carried out. Results: Compared with the control group, body weights in the model group, the Ganmai Dazao Decoction group and the Xiangshao Granule group were decreased(P<0.01). The social isolation coupled with residential invasion could significantly increase horizontal movement and upright numbers in the open field test, and increase the attack numbers, prolong the attack time, shorten the assault latent period and increase the aggressive behavior score in the aggressive behavior testing(P<0.05, P<0.01). But, the corresponding treatment measures improved the body weight and behavior tests parameters(P<0.05). Notably, there were no statistical differences in these index between the Ganmai Dazao Decoction group and Xiangshao Granule group. Conclusion: Ganmai Dazao Decoction could affect behavioral characteristics in rat model of ‘anger' and regulate ‘anger' behavior.
