HPLC fingerprint and quality evaluation of apricot flower from different producing areas

摘要 目的:建立杏花药材(HPLC)指纹图谱测定方法,对不同产地的杏花药材进行质量评价。方法:采用正交设计优化杏花提取方法,建立HPLC指纹图谱。运用相似度评价、聚类分析与主成分分析相结合的方式对不同产地杏花做出统计分析及质量评价。结果:正交设计最佳提取工艺为70%乙醇20m L超声30min。10批样品共确定15个共有峰,相似度均>0.900,指认了3个特征峰,分别为绿原酸、芦丁、阿魏酸。聚类分析将10批药材聚为4类;主成分分析提取出3个特征峰,其累计贡献率为87.525%。根据3个主成分的综合评价得分判定:10批次样品中来自河北沽源、北京、河北张家口3个产地的山杏花综合得分最高,质量最好。结论:不同产地与基源的杏花药材化学成分相似,但含量存在显著性差异,山杏花较杏花质量好。聚类分析和主成分分析相互补充,互为佐证,与相似度评价三者相结合为杏花药材的质量控制提供依据。 Objective: To establish HPLC fingerprint determination method of apricot flower and evaluate the quality of apricot flower from different producing areas. Methods: Extraction method was optimized by orthogonal experiment design and the HPLC fingerprints were defined. The statistical analysis and quality evaluation of apricot flowers from different areas were made by the combination of the similarity evaluation, clustering analysis and principal component analysis. Results: The optimum conditions of extraction process were as follows: 70% ethanol, 30 minutes, 20 m L. Fifteen common peaks were detected in 10 batches of apricot flower and the similarities were all above 0.900. Three peaks were verified including chlorogenic acid, rutin and ferulic acid. Ten batches of medicines were divided into 4 classes according to clustering analysis. Three principal components were extracted, whose cumulative contribution rate was 87.525%. According to the comprehensive scores of 3 principal components, the quality of mountain apricot flower from Guyuan Hebei, Beijing and Zhangjiakou Hebei were the best, which got the highest scores. Conclusion: The chemical composition of apricot flowers from different producing areas is similar, but the content shows a significant difference. The quality of mountain apricot flower is better than apricot flower. Cluster analysis and principal component analysis complements and supports each other. Combining with the similarity evaluation, it provides the basis for quality control of apricot flower.
