摘要
目的:观察柴芪汤对非酒精性脂肪性肝病(NAFLD)大鼠肝脏肝X受体α(LXRα)蛋白表达的影响,探讨其防治NAFLD的可能机制。方法:将32只SPF级雄性SD大鼠随机分为正常组、模型组、柴芪汤组和罗格列酮组,每组8只,采用特殊高脂饲料喂养8周复制NAFLD模型,造模第1天开始用药组分别给予柴芪汤(5.67g·kg^(-1)·d^(-1))及罗格列酮混悬液(3mg·kg^(-1)·d^(-1))灌胃干预,8周后检测大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、甘油三酯(TG)、胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C);观察肝组织病理形态改变;Western blot及免疫组化法测定肝LXRα蛋白表达。结果:模型组大鼠血清ALT、AST、TC、TG、LDL-C水平明显高于正常组(P<0.05),HDL-C水平明显低于正常组(P<0.05),两用药组各项指标优于模型组(P<0.05)。柴芪汤可更明显的降低肝脏组织中LXRα蛋白的表达(P<0.05)。结论:柴芪汤能够改善NAFLD大鼠肝脏脂肪变,抑制LXRα蛋白表达,降低NAFLD大鼠血清ALT、AST、TG、TC、LDL-C,提高HDL-C,这可能是柴芪汤治疗NAFLD的机制之一。
Objective: To observe the effects of Chaiqi Decoction on the protein expression of LXRα in the livertissue of non-alcoholic fatty liver disease(NAFLD) rats, and to investigate the possible mechanism of prevention and treatment of NAFLD. Methods: Thirty-two SPF male SD rats were randomly divided into normal group, model group, Chaiqi Decoction group and rosiglitazone group, with eight rats in each group. The rat model was established by feeding high fat diet for 8weeks. The Chaiqi Decoction group was given Chaiqi Decoction(5.67 g ·kg^(-1)·d^(-1)) from the beginning of modeling, and rosiglitazone group was given rosiglitazone(3mg ·kg^(-1)·d^(-1)). The ALT, AST, TG, TC, HDL-C, LDL-C in blood were detected after 8 weeks. The pathological changes of liver tissue were observed. The protein expression of LXRα in liver tissue was detected by Western Blot and immunohistochemistry. Results: Compared with the normal group, the ALT, AST, TG, TC, LDL-C in blood serum were significantly higher in model group(P<0.05), and HDL-C in blood serum were significantly lower in model group(P<0.05). All the indexes of the treatment group were lower than those of the model group(P<0.05). The protein expression levels of LXRαin the treatment groups were decreased compared with the model group, and Chaiqi Decoction group had better result than the rosiglitazone group(P<0.05). Conclusion: Chaiqi Decoction can improve the hepatic steatosis, inhibit the protein expression of LXRα and decrease the serum levels of ALT, AST, TG, TC, LDL-C and increase the serum levels of HDL-C in NAFLD rats, which may be one of the mechanisms of Chaiqi Decoction in the treatment of NAFLD.
