接骨膏对家兔骨折愈合过程中BMPR-Ⅱ及SMAD8 mRNA表达的影响 被引量:2

Effects of Jiegu Onitment on BMPR-Ⅱ and SMAD8 mRNA expression during fracture healing process of rabbits

摘要 目的:探讨接骨膏外敷给药对家兔骨折愈合过程中BMPR-Ⅱ、SMAD8 m RNA表达的影响,分析其促进骨折愈合的作用机制。方法:建立30只家兔桡骨骨折模型,随机分为实验组和对照组,每组15只,实验组在其患处外敷接骨膏并用小夹板固定;对照组不使用药物,仅常规消毒后进行小夹板固定。分别于给药后第3、6、10周每组随机抽取5只,处死家兔取患处组织标本,采用实时荧光定量PCR方法测定家兔骨组织中BMPR-Ⅱ、SMAD8m RNA的含量,对所得数据进行统计学分析。结果:实验组家兔第3、10周末患处骨组织标本中BMPR-Ⅱ、SMAD8m RNA表达均高于对照组(P<0.05)。结论:接骨膏可在骨折愈合早、晚期促进患处骨组织中BMPR-Ⅱ、Smad8m RNA的表达,加速骨折愈合。 Objective: To investigate the effects of external application of Jiegu Onitment on the expression of BMPRI and SMAD8 m RNA in the fracture healing process of rabbit, and to analyze the underlying mechanism. Methods: The rabbit model of radius fracture was established and then the rats were randomly divided into the experimental group and the control group, 15 rabbits per group. The experimental group was treated with small splint fixation combined with external application of Jiegu Onitment, and the control group was treated with only small splint fixation. Five rabbits were randomly selected from each group after 3, 6 and 10 weeks administration, and then the rabbits were sacrificed and the bone tissue was harvested for BMPRI and SMAD8 m RNA detection by real-time fluorescence quantitative PCR method. Results: Real-time fluorescence quantitative PCR results showed that the BMPR-I and SMAD8 m RNA expression of the experimental group at the 3rd and the 10 th week were higher than those of the control group(P<0.05). Conclusion: Jiegu Onitment could accelerate fracture healing process by promoting BMPR-I and SMAD8 m RNA expression of the bone tissue at early and late period.
