摘要
目的:探讨南蛇藤提取物对人食管鳞癌TE-8细胞凋亡的影响,并对其机制进行初步探讨。方法:MTT法检测不同质量浓度的南蛇藤提取物作用于TE-8细胞24、48h后细胞的增殖情况。南蛇藤提取物作用TE-8细胞24h后,Annexin V-FITC/PI双染法检测细胞凋亡情况;JC-1试剂盒检测线粒体膜电位的损伤;TUNEL法检测DNA损伤情况;Western blot法检测丝裂原活化的蛋白激酶(MAPK)信号通路相关蛋白的表达情况。结果:南蛇藤提取物能明显抑制人食管鳞癌TE-8细胞的增殖,呈时间及浓度依赖;南蛇藤提取物能促进细胞凋亡,降低线粒体膜电位,诱导细胞内DNA损伤,阻断MAPK信号通路的表达,呈浓度依赖。结论:南蛇藤提取物能够抑制TE-8细胞的增殖,促进其凋亡,其机制可能与南蛇藤提取物抑制MAPK信号通路的表达有关。
Objective: To study the effects and mechanism of celastrus orbiculatus extract(COE) on apoptosis of human esophageal squamous carcinoma TE-8 cells. Methods: The proliferation rates of TE-8 cells treated with different concentrations of COE for 24 and 48 hours were measured by MTT assay. After being treated with COE for 24 hours. The apoptosis rate was examined by the Annexin V-FITC/PI staining. The mitochondria membrane potential was examined according to the JC-1 kit. The DNA damage were measured by TUNEL apoptosis assay Kit. The protein expressions of MAPK signal pathway was detected by Western blot. Results: COE suppressed the proliferation of TE-8 cells in a concentration-and time-dependent manner. COE induced apoptosis, reduced the mitochondria membrane potential, increased DNA damage, blocked the expressions of MAPK pathway in a concertration-dependent manner. Conclusion: COE could inhibit the proliferation and induce the apoptosis, and its mechanism may be related to regulating the expressions of MAPK signal pathway
