增液润燥汤对干燥综合征NOD小鼠颌下腺中T、B淋巴细胞浸润及AQP5表达水平的影响 被引量:4

Effects of Zengye Runzao Decoction on submaxillary gland lymphocyte infiltration and the expression of AQP5 in NOD mice with Sjogren's syndrome

摘要 目的:观察增液润燥汤对干燥综合征(SS)NOD小鼠颌下腺中T、B淋巴细胞浸润情况以及AQP5表达水平的影响。方法:将40只8周龄NOD小鼠随机分为5组,即模型组,增液润燥汤低、中、高剂量组与羟氯喹组,同时选取8只同龄Balb/c小鼠作为正常组。增液润燥汤低、中、高剂量组分别给予10.5、21.0、42.0g/kg增液润燥汤灌胃;羟氯喹组给予60mg/kg羟氯喹灌胃;正常组与模型组给予同等剂量的去离子水灌胃。8周后,测量体质量后于麻醉下处死,摘取颌下腺、脾腺与胸腺称重并计算脏器指数;使用免疫组化法检测小鼠颌下腺中T、B淋巴细胞浸润量以及水通道蛋白5(AQP5)表达水平。结果:模型组小鼠颌下腺指数与正常组比较明显下降(P<0.05),药物干预后小鼠颌下腺指数变化不明显。模型组小鼠脾脏和胸腺指数与正常比较变化不明显。免疫组化结果显示,正常组小鼠颌下腺中未见有T与B淋巴细胞的浸润,模型组小鼠颌下腺中可见有大量的T、B淋巴细胞浸润;干预8周后,增液润燥汤各剂量组小鼠颌下腺中T、B淋巴细胞浸润量与模型组比较皆明显下降(P<0.05)。模型组小鼠AQP5的表达水平与正常组小鼠比较明显降低(P<0.05);干预8周后,增液润燥汤各剂量组小鼠颌下腺中AQP5的表达水平皆明显上升(P<0.05)。结论:NOD小鼠颌下腺中T和B淋巴细胞的浸润量明显增多、AQP5的表达明显减少;增液润燥汤能够有效抑制NOD小鼠颌下腺中T和B淋巴细胞浸润、促进AQP5的表达。 Objective: To observe the effects of Zengye Runzao Decoction on the infiltration of T, B lymphocyte in submaxillary glands and the expression of AQP5 in submaxillary glands of non-obese diabetic(NOD) mice with Sj?gren's syndrome(SS). Methods: Forty NOD mice were randomly divided into 5 groups: model group, low, middle, and high dosage of Zengye Runzao Decoction groups, and hydroxychloroquine group, and meanwhile, eight Balb/c mice were selected as normal control group. The low, middle, high dosage of Zengye Runzao Decoction groups received gavage administration with 10.5, 21, 42g/kg Zengye Runzao Decoction respectively and the hydroxychloroquine group received gavage administration with 60g/kg hydroxychloroquine. The normal and model groups received gavage administration with pure water. After 8 weeks, bilateral submaxillary glands, spleen and thymus were weighted to calculate organ indexesunder anesthesia. The infiltration of T, B lymphocyte and expression of AQP5 in submaxillary glands were examined through immunohistochemistry. Results: Compared with the normal group, the submaxillary glands index of mouse in model group were reduced significantly(P<0.05) and the Zengye Runzao Decoction had no obviously effect on it. Immunohistochemical results showed that there was no infiltration of T and B lymphocytes in the submandibular gland of normal control group but lots of infiltration of T and B lymphocytes in the submandibular gland of model group. Compared with the model group, the infiltration of T and B lymphocytes in the submandibular gland in low, middle, and high dosage of Zengye Runzao Decoction groups were reduced significantly(P<0.05). Compared with normal control group the AQP5 expression in the submandibular gland were reduced significantly in the model group(P<0.05), after intervening for 8 weeks, AQP5 expression in the submandibular gland significantly increased of low, middle, and high dosage of Zengye Runzao Decoction groups(P<0.05). Conclusion: The infiltration of T and B lymphocyte in submaxillary gland of NOD mice is increased significantly. The expression of AQP5 in submaxillary glandof NOD mice is decreased significantly. And Zengye Runzao Decoctioncan effectively inhibit the infiltration of T and B lymphocyte and promote the expression of AQP5.
