摘要
目的:研究组分中药KX组合物及其等剂量的单味组分对实验性自身免疫性心肌炎(EAM)模型的影响。方法:制备EAM模型,于首次免疫后2、8周,分别测定血液中IL-2、IL-17、TNF-α、IFN-γ的含量,取心脏做病理检查。结果:注射抗原2周后,模型大鼠血液中IL-2、IL-17、TNF-α、IFN-γ明显升高(P<0.05,P<0.01)。8周时IFN-γ持续升高(P<0.05),其余因子水平恢复正常。心肌由2周的细胞变性、炎细胞浸润逐渐演变为组织纤维化、坏死。苦参总碱给药2周时降低血液中IL-17、TNF-α、IFN-γ(P<0.01);8周时IL-17、TNF-α持续降低(P<0.01,P<0.05)。西洋参总皂苷2周时降低了IL-2、IL-17、TNF-α、IFN-γ,8周时IL-2、IL-17、IFN-γ仍降低。KX组合物2周时降低了IL-2、IL-17、TNF-α、IFN-γ(P<0.01,P<0.05),并持续到8周(P<0.01)。病理结果表明,各给药组心肌损伤程度均轻于模型组。结论:KX组合物及其单味组分能不同程度的明显抑制EAM模型Th1、Th17细胞亚群激活的相关细胞因子的释放,减轻心肌损伤;组合物的作用效果体现了中药的配伍优势。
Objective: To study the effects of KX composition and single composition in equivalent dose on the autoimmunity myocarditis(EAM) model. Methods: The model of EAM was established. After 2 weeks and 8 weeks by the first immunization, the content of IL-2, IL-17, TNF-α and IFN-γ in serum were measured, and the hearts weretaken out for pathological examination. Results: IL-2, IL-17, TNF-α IFN-γ in the serum of model rats were significantly increased 2 weekslater afterthe injectionof antigen in model animals. In the 8th week, the content of IFN-γcontinued to rise while the level of other factors returned to normal. Myocardium was from the cellular degeneration and infiltration of inflammatory cells in the 2nd week to fibrosis and necrosis. Total alkaloids reduced the content of IL-17, TNF-α and IFN-γ in the 2nd week and the content of IL-17, TNF-αcontinued to reduce in the 8th week. The total saponins of American ginseng decreased the content of IL-2, IL-17, TNF-α and IFN-γ in the 2nd week. The content of IL-2, IL-17 and IFN-γ continued to decrease in the 8th week. KX composition reduced the content of IL-2, IL-17, TNF-α and IFN-γ, and continued to reduce until the 8th week. Pathology determination found that the injury degree of myocardium in medication administration groups were lighter than that in model group. Conclusion: The KX composition and single composition could inhibit release of cytokines due to Th1 and Th17 cell subsets activated, reduce myocardial injury of EAM model. The effects of the composition give expression to the compatibility advantages of traditional Chinese medicine.
