抵挡汤早期干预对2型糖尿病大鼠血管内皮细胞凋亡的影响 被引量:9

摘要 目的:探讨抵挡汤早期干预对2型糖尿病大鼠血管内皮细胞凋亡的影响。方法:将SD大鼠随机分为正常对照组,糖尿病模型组,抵挡汤早、中、晚期干预组,二甲双胍组,辛伐他汀组。酶联免疫吸附实验(ELISA)检测活性氧(ROS)含量,实时荧光定量PCR检测凋亡诱导因子(AIF)、凋亡蛋白激酶激活因子-1(Apaf-1)、半胱天冬肽酶-3(Caspase-3)基因表达水平。结果:与正常对照组比较,糖尿病模型组ROS含量,AIF、Apaf-1、Caspase-3基因表达水平均显著升高(P<0.05);与糖尿病模型组比较,抵挡汤早、中期干预组,二甲双胍组及辛伐他汀组ROS含量,AIF、Apaf-1、Caspase-3基因表达水平均显著下降(P<0.05),且在抵挡汤早期干预组中AIF、Apaf-1、Caspase-3基因表达水平显著低于中、晚期干预组(P<0.05)。结论:抵挡汤早期干预可以通过抗氧化应激减少血管内皮细胞凋亡。 Objective: To Explore the effects of Didang Decoction(DDD) early intervention on vascular endothelial cell apoptosis in type 2 diabetic rats. Methods: SD rats were randomly divided into seven groups: normal control group, diabetes model group, DDD early-stage intervention group, middle-stage intervention group, late-stage intervention group, metformin group and simvastatin group. The content of ROS was detected by ELISA. The gene expression levels of AIF, Apaf-1 and Caspase-3 were measured by real-time quantitative PCR. Results: Compared with normal control group, the levels of ROS, AIF, Apaf-1 and Caspase-3 were significantly up-regulated in diabetes group(P<0.05). Compared with diabetes group, the levels of ROS, AIF, Apaf-1 and Caspase-3 were significantly down-regulated in DDD early/middle-stage intervention groups, metformin group and simvastatin group(P<0.05). The gene expression levels of AIF, Apaf-1 and Caspase-3 in DDD early-stage intervention group were significantly lower than those in DDD-middle stage intervention group(P<0.05). Conclusion: DDD early-stage intervention can inhibit vascular endothelial cell apoptosis by anti-oxidative stress.
