朝医麻黄定喘汤对哮喘小鼠NF-κB信号通路的影响 被引量:10

Effects of Mahuang Dingchuan Decoction from traditional Korean medicine on NF-κB signal pathway in asthmatic mice

摘要 目的:探讨朝医麻黄定喘汤对支气管哮喘小鼠气道重塑及核因子-κB(NF-κB)信号通路的影响。方法:50只清洁级雌性BABL/c小鼠,随机分为5组,每组10只,分别为正常对照组,模型组,麻黄定喘汤低、高剂量组,地塞米松组。以卵清蛋白致敏、激发制备小鼠哮喘模型,在末次激发24h后处死。取小鼠肺泡灌洗液(BALF)及肺组织检测炎性细胞的数量变化,白介素-4(IL-4)、白介素-5(IL-5)、白介素-13(IL-13)的水平及NF-κBp65蛋白的表达。结果:与正常对照组比较,模型组BALF中炎性细胞计数、IL-4、IL-5、IL-13水平均增高;肺组织NF-κBp65蛋白表达水平均显著升高(P<0.01)。与模型组比较,麻黄定喘汤组小鼠BALF中炎性细胞计数、IL-4、IL-5、IL-13水平及NF-κBp65蛋白表达水平均显著降低(P<0.05,P<0.01),不同剂量麻黄定喘汤组间上述各指标间差异无统计学意义。结论:麻黄定喘汤可抑制哮喘小鼠气道重塑的发生,其机制可能是通过抑制NF-κB信号通路而实现的。 Objective: To investigate the effects of Mahuang Dingchuan Decoction(MHDCD) from traditional Korean medicine on the airway remodeling and nuclear factor-κB(NF-κB) signaling pathway in asthmatic mice. Methods: Fifty female BABL/c mice of clean grade were randomly divided into five groups(n=10): control group, model group, low dose of Mahuang Dingchuan Decoction group, high dose of Mahuang Dingchuan Decoction group, dexamethasone group. Mouse model of asthma were sensitized and challenged with Ovalbumin(OVA), the left lung of the mice were stained with hematoxylin-eosin(HE) staining, PAS staining and Masson staining after 24 h of the last challenge. The right lung tissues were detected by enzyme-linked immunosorbent assay(ELISA) to observe the expression levels of interleukin-4(IL-4), interleukin-5(IL-5) and interleukin-13(IL-13) in bronchoalveolar lavage fluid(BALF), the protein expression levels of NF-κBp65 was detected by RT-PCR and Western blot. Results: The inflammatory cell counts and the expression levels of IL-4, IL-5 and IL-13 in BALF were significantly increased in the model group compared with the control group(P<0.01). The expression levels of NF-κBp65 protein in the lung tissue of the model group was significantly higher than that in the control group(P<0.01). The inflammatory cell counts and the expression levels of IL-4, IL-5, IL-13 and NF-κBp65 protein levels in BALF of mice in Mahuang Dingchuan Decoction group were significantly lower than those of model group(P<0.05, P<0.01), there was no significant difference in the above-mentioned indexes between low dose of Mahuang Dingchuan Decoction group and high dose of Mahuang Dingchuan Decoction group. Conclusion: Mahuang Dingchuan Decoction can inhibit the airway remodeling in asthmatic mice, part of the mechanism may be achieved through the inhibition of NF-κB signaling pathway.
