白豆蔻挥发油对糖尿病肾病大鼠肾脏保护作用 被引量:21

Protective effect of Amomum kravanh volatile oil on kidney in diabetic nephropathy rats

摘要 目的:研究白豆蔻挥发油对糖尿病肾病大鼠肾脏保护作用及其机制。方法:46只Wistar大鼠随机分为正常组(6只)和模型组(40只),模型组以腹腔注射STZ建立糖尿病肾病模型,造模后随机分模型组,白豆蔻低、高剂量组,六味地黄丸组4组,各10只。连续灌胃给药21d,每周检测1次血糖;最后一次给药24h后麻醉大鼠腹主动脉取血,检测血清肌酐(Cre)、尿素氮(BUN)、白蛋白(ALB)含量;光镜下观察肾脏组织病理改变;免疫组化法检测基质金属蛋白酶2(MMP-2)、转化生长因子β1(TGF-β1)、胰岛素样生长因子2(IGF-2)的表达。结果:与正常组比较,模型组血糖显著升高(P<0.01),经过给药后各给药组血糖不同程度下降,其中白豆蔻高剂量组血糖显著下降(P<0.05,P<0.01);与模型组比较,各给药组Cre、BUN、ALB含量均降低,其中白豆蔻高剂量组作用明显(P<0.05,P<0.01)。光镜下病理形态学观察显示,各给药组对大鼠肾组织损伤有不同程度的改善作用;各给药组MMP-2、TGF-β1、IGF-2蛋白表达均上调。结论:白豆蔻挥发油对肾脏有一定的保护作用,作用机制可能与上调MMP-2、TGF-β1、IGF-2的表达有关,从而改善链脲佐菌素所致糖尿病肾病的病理改变。 Objective: To study the protective effect of volatile oil of Amomum kravanh onkidney of diabetic nephropathy rats and its mechanism. Methods: Forty-six Wistar rats were randomly divided into normal group(n=6) and model group(n=40). The model group was injected intraperitoneally with streptozotocin(STZ) to establish diabetic nephropathy model. After modeling, the rats were randomly divided into model group, low dose of Amomum kravanh volatile oil group, high dose of Amomum kravanh volatile oil group and Liuwei Dihuang Pill group(n=10 in each group). Continuous intragastric administration was given for 21 days and blood glucose was tested weekly. After 24 h of the last administration, rats were sacrificed to take blood from abdominal aorta, and the levels of serum creatinine(CRE), blood urea nitrogen(BUN) and albumin(ALB) were tested. The pathological changes of renal tissue were observed under light microscope. The expressions of MMP-2, TGF-β1, and IGF-2 were detected by immunohistochemistry. Results: Compared with normal group, the level of blood glucose of model group was increased significantly(P<0.01), and after administration of the medicine, the levels of blood glucose of each group were decreased in varying degrees. In the high dose of Amomum kravanh volatile oil group, the blood glucose was lower than those of the low dose of Amomum kravanh volatile oil group and Liuwei Dihuang Pill group(P<0.05, P<0.01). Compared with the model group, the levels of Cre, BUN and ALB in each administration group were decreased, and the effects of high dose group was significant(P<0.05, P<0.01). Pathological and morphological observation under light microscope showed that the rats in each administration group had different improvement effects on the renal tissue injury. The expressions of MMP-2, TGF-β1 and IGF-2 in renal tissue were up-regulated in all administration groups. Conclusion: Amomum kravanh volatile oil has a certain protective effect on the kidney, and the mechanism may be related to up-regulating the expressions of MMP-2, TGF-β1 and IGF-2, so as to improve the pathological changes of diabetic nephropathy induced by STZ.
