藏药材全缘叶绿绒蒿总黄酮的富集纯化工艺研究 被引量:8

Enrichment and purification of total flavonoids of Meconopsis integrifolia(Maxim.) Franch in Tibetan herbs with macroporous resins

摘要 目的:研究大孔吸附树脂法富集和纯化藏药材全缘叶绿绒蒿的活性部位总黄酮的工艺参数和条件。方法:以全缘叶绿绒蒿总黄酮为考察指标,对AB-8、D101、HPD450和HPD600 4种不同极性大孔树脂富集纯化总黄酮的吸附和解吸性能进行评价。结果:D101型大孔吸附树脂适用于富集纯化全缘叶绿绒蒿总黄酮,最佳工艺条件为:吸附条件为上样质量浓度0.30g生药/m L,洗脱液体积流量为3BV/h,最佳吸附容量为51.2m L/g;解吸条件为4BV蒸馏水洗脱去除杂质后,换用50%乙醇4BV洗脱。经过D101大孔树脂富集纯化后,全缘叶绿绒蒿总黄酮的纯度由2.47%上升至33.83%,平均回收率91.35%。结论:D101型大孔吸附树脂对全缘叶绿绒蒿总黄酮有良好的吸附及解吸性能,能有效富集和纯化全缘叶绿绒蒿总黄酮,可为全缘叶绿绒蒿有效活性总黄酮部位的制备提供参考依据。 Objective: To study the technological parameter and condition for enrichment and purification of total flavonoids of Meconopsis integrifolia(Maxim.) Franch by macroporous resins. Methods: With the content of Meconopsis integrifolia(Maxim.) Franch total flavonoids as index, static and dynamic adsorption-desorption of AB-8, D101, HPD450 and HPD600 were investigated to enrich and purify total flavonoids. Results: D101 macroporous resin was selected for enrichment and purification of total flavonoids. The optimal condition was sample concentration 0.30g/m L, elution flow rate 3BV/h, and adsorption capacity 51.2m L/g, and sample was first eluted with 4BV of water, then with 3BV 50% ethanol. The purity of total flavonoids was increased from 2.47% to 33.83%. Conclusion: The total flavonoids of Meconopsis integrifolia(Maxim.) Franch was enrichment and purification effectively for the good adsorption-desorption effect of D101 macroporous resin. This method could be used for preparing active site of Meconopsis integrifolia(Maxim.) Franch.
