清化固肾排毒颗粒对单侧输尿管梗阻大鼠肾组织NF-κB、MCP-1表达的影响 被引量:5

Effects of Qinghua Gushen Paidu Granules on expressions of NF-κB and MCP-1 in nephrogenic tissue of rats with unilateral ureteral obstruction

摘要 目的:观察清化固肾排毒颗粒(QGPG)对单侧输尿管梗阻大鼠NF-κB、MCP-1表达的影响并探讨其作用机制。方法:108只SD大鼠随机分为假手术组,模型组,洛汀新(1.60mg/kg)组和中药高、中、低剂量(3.00、1.50、0.75g/kg)组,每组18只,除假手术组外,各组大鼠均以结扎左侧输尿管造模。各组分别于造模第7、14、21天时随机处死6只大鼠,取肾组织Masson染色观察肾纤维化,免疫组化检测肾组织NF-κB、MCP-1表达。结果:与假手术组比较,模型组肾纤维化明显(P<0.05),肾组织NF-κB、MCP-1表达显著增多(P<0.05);中药各剂量组和洛汀新组较模型组上述指标均显著性下降(P<0.05);与洛汀新组比较,中药各剂量组NF-κB、MCP-1表达无统计学差异;中药各剂量组组间比较,肾纤维化和NF-κB、MCP-1表达无统计学差异。结论:QGPG可以有效延缓肾纤维化,可能通过下调NF-κB、MCP-1表达,从而抑制炎性反应起到抗纤维化作用。 Objective: To observe the effects of Qinghua Gushen Paidu Granules(QGPG) on expression of NF-κB, MCP-1 in nephrogenic tissue of rats with unilateral ureteral obstruction and investigate the mechanism of it. Methods: The 108 male Sprague-Dawley(SD) rats were randomly divided into Sham-operated group, model group, Lotensin treated group(1.6mg/kg) and high, middle, low dosage of Chinese medicine groups(3.00, 1.50 and 0.75g/kg), 18 rats per each group. Except for the Shamoperated group, the model was established by ligating left ureter. At 7th, 14 th and 21 st in model establishment, six rats in each group were randomly sacrificed, and their nephrogenic tissue were collected for observing renal fibrosis by Masson staining and the expression of NF-κB and MCP-1 by immunohistochemical method. Results: Compared with the Sham-operation group, the renal fibrosis degree was increased in the model group(P<0.05) and the protein expression of NF-κB, MCP-1 was significantly increased(P<0.05), all of which were reversed in the high, middle, low dosage of Chinese medicine groups as well as the Lotensin treated group(P<0.05). The protein expressions of NF-κB, MCP-1 in high, mid, low dosage Chinese medicine groups were not different significantly compared with the Lotensin treated group. Similarly, there were no significant difference in the renal interstitial fibrosis degree and the protein expression of NF-κB, MCP-1 among the high, mid, low dosage Chinese medicine groups. Conclusion: QGPG may effectively retard renal fibrosis by reducing the protein expression of NF-κB, MCP-1 to inhibit the inflammatory reaction.
