益气活血方对慢性心力衰竭大鼠肺组织水通道蛋白表达的影响 被引量:10

Effects of Yiqi Huoxue Formula on the expression of AQP1 in the lung tissue of chronic heart failure rats

摘要 目的:通过观察益气、活血代表方保元汤联合桃红四物汤对心肌梗死后心力衰竭大鼠肺组织水通道蛋白1(AQP1)表达的影响,探讨益气活血方改善慢性心力衰竭肺泡水转运的作用机制。方法:结扎左冠状动脉前降支建立大鼠急性心肌梗死(AMI)模型,术后4周根据超声心动图结果随机将动物分为模型组、益气活血组和缬沙坦组,并另设假手术组。用药4周后,观察大鼠肺组织病理学改变及肺/体重比,通过Western blot检测大鼠肺脏AQP1蛋白表达。结果:与假手术组比较,模型组大鼠肺间质破坏严重,湿肺/体质量比、干肺/体质量比明显升高(P<0.05),肺组织AQP1表达显著下调(P<0.05);与模型组比较,益气活血组和缬沙坦组大鼠肺间质破坏显著减轻,湿肺/体质量比、干肺/体质量比显著降低(P<0.05),肺组织AQP1表达显著上调(P<0.05)。结论:益气活血方能够减轻慢性心力衰竭肺间质损害,调控肺组织AQP1表达改善肺泡水转运,从而改善心脏功能。 Objective: To explore the mechanism of Yiqi Huoxue Formula improve pulmonary alveoli water transport in the lung of chronic heart failure rats through observing the effect of Baoyuan Decoction combined with Taohong Siwu Decoction which are Yiqi Huoxue representative prescription on expression of aquaporin-1(AQP1) in the lung tissue of chronic heart failure rats. Methods: Acute myocardial infarction model in rats was established by ligaturing the proximal left anterior descending branch(LAD) of coronary artery and the rats were randomly divided into four groups as model group, Yiqi Huoxue group, valsartan group and sham-operated group at 4 weeks after operation according to the results of echocardiography survivors. After drug intervention 4 weeks, observing the changes of pathological for lung tissue, lung and body weight ratio in rats and the expression of AQP1 was detected by the method of western blotting. Results: Compared with the sham-operated group, the pulmonary interstitium of model group was severely damaged, and both of the wet lung and body weight ratio and dry lung and body weight ratio were significantly increased(P<0.05), the rats lung AQP1 expression levels was significantly reduced(P<0.05) in model group; Compared with the model group, the pulmonary interstitial damage in Yiqi Huoxue group and valsartan group were reduced, and the wet lung and body weight ratio and dry lung and body weight ratio were significantly lower(P<0.05), and expression levels of AQP1 were significantly increased(P<0.05). Conclusion: Yiqi Huoxue Formula can reduce the lung damage of chronic heart failure and regulate the expression of AQP1 to improve pulmonary alveoli water transport in the lung of rats, which will improve the cardiac function.
