黄芪及其拆分组分对脾虚水湿不化大鼠矿物质吸收相关基因表达谱的影响 被引量:6

Effects of Radix Astragali and its components on gene expression profiles related to mineral absorption in rats with syndrome of dampness stagnancy due to spleen deficiency

摘要 目的:研究黄芪及其拆分组分(黄酮、皂苷、多糖、水溶液)对脾虚水湿不化大鼠十二指肠矿物质吸收相关基因表达谱的影响,阐明黄芪性味归属。方法:高脂低蛋白饮食与力竭游泳法建立脾虚水湿不化大鼠模型,黄芪及其拆分组分灌胃2周。Agilent大鼠基因表达谱芯片检测各组大鼠十二指肠基因表达谱变化,筛选差异基因,并进行生物信息学分析。荧光实时定量PCR验证Trpm6、Reg1α基因表达。结果:与模型组比较,黄芪水煎液组688个差异基因,GO功能分类涉及金属离子转运、阳离子平衡等;黄酮组1 204个差异基因,涉及钠、钾、铁、铜等离子运输;皂苷组1 361个差异基因,涉及二价金属离子外流、铁离子平衡、铜离子解毒等;多糖组3 181个差异基因,涉及阳离子运输、铁离子运输等;水溶液组2 419个差异基因,涉及镁离子应答、阳离子运输等。KEGG分析结果显示,各给药组生物学通路均涉及矿物质吸收,据差异基因富集由高到低为多糖组>黄酮组>水溶液组>皂苷组>黄芪水煎液组。荧光实时定量PCR验证结果和芯片结果一致。结论:黄芪及其拆分组分通过调节模型大鼠十二指肠与铁、铜、镁、磷等吸收相关基因,维持机体离子平衡,体现其性温、味甘,多糖组分作用更好。 Objective: To investigate the effects of Radix Astragali(RA) and its components on gene expression profiles related to mineral absorption in rats with syndrome of dampness stagnancy due to spleen deficiency, and to clarify drug properties of components in RA. Methods: Model rats were induced by high fats and low protein diet plus exhaustive swimming methods. And then RA and its components were administered to corresponding groups for two weeks. Differentially expressed genes(DEGs) were tested by the gene chip technology and performed bioinformatic analysis. The expressions of Trpm6, Reg1αwere tested by q RT-PCR. Results: Compared with the model group, 688 DEGs were screened out in the RA decoction group and Gene Ontology(GO) categories indicated the DEGs were associated with divalent metal export, cellular cation homeostasis. One thousand two hundred and four DEGs were screened out in the flavonoid components group and involved in sodium, potassium, iron and copper ion transport. One thousand three hundred and sixty-one DEGs were screened out in the saponin components group and involved in divalent metal export, iron ion homeostasis, detoxification of copper ion. Three thousand one hundred and eighty-one DEGs were screened out in the polysaccharide components group and involved in cation transport, iron ion transmembrane transport. Two thousand four hundred and ninteen DEGs were screened out in the aqueous solution components group and involved in cation transport, in response to magnesium ion. Analysis of kyoto encyclopedia of genes and genomes(KEGG) showed pathway were involved in mineral absorption in each of these groups, and the polysaccharide components group > the flavonoid components group > the aqueous solution components group > the saponin components group > the RA decoction group according to enrichment condition. Results of q RT-PCR were in accord with gene chip results. Conclusion: RA and its components can improve mineral absorption of model rats through regulating genes related to iron, copper, magnesium and phosphorus absorption, which reflectes sweet and warm in property, and the effect of the polysaccharide components group is the best.
