天丝饮对去势大鼠学习记忆的影响及神经营养相关的研究 被引量:2

Effects of Tiansi Decoction on learning and memory function and neurotrophic in ovariectomized rat model with Alzheimer's disease

摘要 目的:研究天丝饮对去势所致老年痴呆模型行为学及神经营养相关的研究。方法:50只SD大鼠随机分为正常组、模型组、假手术组、雌二醇组、天丝饮组。通过双侧去卵巢造成老年痴呆模型,手术1周后,雌二醇组及天丝饮组分别给予雌二醇(0.2mg/m L)、天丝饮(0.35g/kg),灌胃8周。采用Morris水迷宫检测其5d的学习行为能力,Western blot测试其海马组织中p-p38 MAPK,p-CREB,BDNF的含量。结果:在Morris水迷宫中,与模型组比较,天丝饮组潜伏距离缩短(P<0.05),穿越平台次数明显增多(P<0.01)。Western blot表明,与模型组比较,天丝饮组能降低大鼠大脑海马区p-p38 MAPK的表达,升高p-CREB、BDNF的表达(P<0.01)。结论:天丝饮在一定程度上通过抑制大鼠大脑海马神经区神经凋亡和增强神经营养,改善去势大鼠的学习记忆能力。 Objective: To investigate the effects of Tiansi Decoction on the behaviors and neurotrophic in ovariectomized rats. Methods: Fifty SD rats were randomly divided into normal group, model group, shame group, estradiol group and Tiansi Decoction(TSY) group. Alzheimer's disease model was established by bilateral ovariectomy. One week after surgery, estradiol group and TSY group were administered with estradiol(0.2 mg/m L) and Tiansi Liquid(3.5 g/kg) by gavage, respectively for 8 consecutive weeks. After 5 day's behavioral observation by Morris water maze, specimen were collected from the hippocampal area. The p-p38, p-CREB and BDNF expression were tested by Western blot. Results: The latent route of TSY group in the Morris water maze was significantly shorten(P<0.05), while numbers of crossing platform were markedly increased(P<0.01), compared with model group. The results of Western blot showed that TSY could decrease the protein expression of p-p38 MAPK, and increase the protein expressions of p-CREB and BDNF in the rat hippocampus. Conclusion: Tiansi Decoction could inhibite the rat brain hippocampal neural apoptosis and enhance the neurotrophic area to a certain extent, improve the ability of learning and memory in ovariectomized rats.
