电针足少阳经穴对去卵巢大鼠骨质疏松症骨组织OPG、RANKL及CBFα1 mRNA表达的影响 被引量:25

Effects of electroacupuncture on foot Shaoyang meridian points on m RNA expressions of POG, RANKL and CBFα1 in bone tissue of ovariectomized osteoporosis rats

摘要 目的:探讨电针足少阳经穴对去卵巢大鼠骨质疏松症(OP)治疗作用的分子学机制。方法:将40只Wistar雌性大鼠随机分为假手术组、模型对照组、模型加胆经针刺组(简称胆经组)、模型加非经穴针刺组(简称非经穴组)。造模3个月后进行不同的干预,6个疗程后后处死。取左侧股骨置冻存管,放-70℃冰箱待测。结果:胆经组OPG mRNA较模型对照组和假手术组显著降低(P<0.01),RANKL mRNA较模型对照组明显降低(P<0.01),OPG/RANKL mRNA的比值较假手术组明显降低(P<0.01),CBFα1 mRNA表达明显高于假手术组(P<0.01)。结论:电针足少阳经穴具有抑制骨吸收同时刺激骨形成的作用,通过抑制破骨细胞关键调控因子OPG和RANKL mRNA表达,提高OPG/RANKL两者比值,抑制骨吸收;同时刺激成骨细胞骨形成特异性基因CBFα1 mRNA表达,增强骨形成,以促进成骨细胞和破骨细胞相偶联,进而达到调骨重建的作用。 Objective: To explore the molecular mechanisms of electroacupuncture(EA) on foot Shaoyang meridian in treating ovariectomized osteoporosis rats. Methods: Forty female Wistar rats were randomly divided into four groups: sham operation group, control group, gall bladder(GB) meridian group and non-meridian group. Three months after ovariectomized surgery, different interventions were applied in each group, and rats were sacrificed six months after ovariectomized surgery.The left femur was taken out quickly, and kept in-70℃. Results: In GB group, the mRNA expression of OPG was significantly lower than that of sham operation group and control group(P<0.01). The mRNA expression of RANKL in GB group was also significantly lower than that of control group(P<0.01). The ratio of mRNA of OPG/RANKL in GB group was decreased than that of sham operation group, and the difference was statistically significant(P<0.01). The mRNA expression of CBFα1 in GB group was significantly higher than that in sham operation group(P<0.01). Conclusion: EA on foot Shaoyang meridian can inhibit bone resorption and promote bone formation. Bone resorption is inhibited by inhibiting the mRNA expressions of OPG and RANKL in osteoclasts and increasing the ratio of OPG/RANKL, while bone formation is promoted by increasing the mRNA expression of specific gene CBFα1, which promotes OB and OC's phase coupling and benefits bone's remodeling.
