摘要
目的:建立灵芝三萜类化合物的高效液相色谱(HPLC)和薄层色谱(TLC)标准指纹图谱。方法:优化洗脱方法、色谱柱、流动相体系等HPLC色谱条件,建立10批不同来源灵芝样品的HPLC指纹图谱,采用中药色谱指纹图谱相似度评价系统(2004A版)计算相似度。对TLC展开剂、显色剂和点样量等因素进行优化,利用优化条件对样品进行TLC分析并重叠共有峰。结果:确定灵芝三萜提取物的HPLC图谱中10个主要特征峰,计算10批次灵芝样品的相似度均>90%,建立了灵芝三萜的TLC指纹图谱,得到7个共有峰,聚类分析结果显示重叠效果较好。结论:通过对灵芝三萜的HPLC及TLC指纹图谱分析,为灵芝质量标准的建立及其不同来源的灵芝药材鉴别起到一定作用。
Objective: To establish the high performance liquid chromatographic(HPLC) fingerprint and the thin layer chromatography(TLC) fingerprint of Ganoderma triterpenes. Methods: Optimized HPLC conditions by selecting the factors such as gradient elution, HPLC column, and mobile phase system. After HPLC fingerprints of 10 batches of Ganoderma lucidum samples were established, the similarity assay for the different samples was carried out to evaluate their quality by Chinese material medica fingerprint similarity evaluation system(2004 A edition). Then TLC conditions were optimized, including developing solvent system, chromogenic agent and sample volume. TLC fingerprints of Ganoderma lucidum from different sources were set up under the optimized TLC conditions. Results: Ten main characteristic peaks were selected from HPLC fingerprint, the similarity was calculated about 90%. After that, 7 main characteristic peaks were selected from TLC fingerprint. The result of cluster analysis showed that the overlapping effect of 10 batches samples was better. Conclusion: The fingerprint of Ganoderma Triterpenes plays an important role to establish the quality standard of Ganoderma lucidum and identify different source of medicinal herbs.
