蠲哮汤对哮喘大鼠气道重塑细胞增殖信号通路的影响 被引量:8

Effects of Juanxiao Decoction on regulating cell proliferation signaling pathway of airway remodeling in asthmatic rats

摘要 目的:观察蠲哮汤对哮喘大鼠模型气道重塑细胞增殖信号通路的影响。方法:60只SPF级雌性SD大鼠,随机分为6组:正常组,模型组,地塞米松组,蠲哮汤低、中、高剂量组,每组10只。除正常组外,各组均采用卵蛋白(OVA)联合雾化吸入的方法建立哮喘大鼠模型。采用实时荧光定量PCR检测各组大鼠肺组织中细胞外信号调节激酶(ERK)1、ERK2及磷脂酰肌醇3激酶(PI3K)mRNA的表达水平。结果:与正常组比较,模型组ERK1、ERK2和PI3K mRNA表达上调(P<0.05);与模型组比较,各治疗组大鼠ERK1、ERK2和PI3K mRNA表达下调(P<0.05)。结论:蠲哮汤可以有效抑制哮喘大鼠的ERK1、ERK2和PI3K mRNA表达,达到调控细胞增殖信号通路的目的,从而抑制哮喘大鼠气道重塑的进程。 Objective: To observe the effects of Juanxiao Decoction on cell proliferation signaling pathway of airway remodeling in asthmatic rats.Methods: Sixty female SD rats were randomly divided into 6 groups: normal group,model group,dexamethasone group,low,medium and high dose Juanxiao Decoction group(n=10).Except the normal group,all rats in other groups were established asthma model by using ovalbumin(OVA) combined with aerosol inhalation.The expression levels of ERK1,ERK2 and PI3K in lung tissue were detected by real time fluorescence quantitative PCR.Results: Compared with the normal group,the expressions of ERK1,ERK2 and PI3K mRNA were up-regulated in model group(P<0.05).Compared with the model group,the expression of ERK1,ERK2 and PI3K mRNA in the dexamethasone group and the traditional Chinese medicine group were decreased(P<0.05).Conclusion: Juanxiao Decoction can effectively inhibit the airway remodeling in asthmatic rats by regulation of cell proliferation signaling pathway via inhibiting the expressions of ERK1,ERK2 and PI3K in the lung tissue.
