土荆芥挥发油对小鼠体内幽门螺杆菌清除作用及对NF-κB表达的影响

Study on bactericidal activity against H.pylori in vivo and effects on the NF-κB expression of Chenopodium ambrosioides L.volatile oil

目的:观察土荆芥挥发油对幽门螺杆菌(Hp)感染小鼠的体内Hp清除作用及对胃黏膜组织NF-κB表达水平的影响。方法:44只SPF级昆明小鼠采用经口感染方法建立Hp感染模型,随机分为空白组、模型组、三联组、土荆芥组。空白组不予干预,模型组给予灭菌注射用水,三联组予兰索拉唑+甲硝唑+克拉霉素给药1周,土荆芥组给予土荆芥挥发油4周,所有动物均于给药4周后处死。快速尿素酶试验、组织病理学染色判定Hp根除情况,免疫组织化学方法测定胃黏膜组织NF-κB p65水平。结果:三联组Hp根除率为70%(7/10),土荆芥组为50%(5/10),两组比较差异无统计学意义(Fisher精确概率法P=0.65)。模型组细胞核NF-κB p65平均光密度及细胞核/细胞浆比值较空白组显著升高(P<0.05,P<0.01),治疗后三联组及土荆芥组NF-κB p65有降低趋势,但差异均无统计学意义。结论:土荆芥具有根除Hp的作用,其对胃黏膜组织NF-κB入核过程可能有一定程度的抑制作用。

Objective: To observe the bactericidal activity against H.pylori and effects on the NF-κB expression of Chenopodium ambrosioides L.volatile oil on H.pylori infected mice model.Methods: Forty-four SPF class KM mice were infected by H.pylori through oral inoculation of H.pylori,which were randomly divided into blank group,control group,triple therapy group and Chenopodium ambrosioides L.group.The treatment strategies were control group with sterile water for 4 weeks,triple therapy group with lansoprazole,metronidazole and clarithromycin for 1 week,and Chenopodium ambrosioides L.group with Chenopodium ambrosioides L.volatile oil for 4 weeks.All the subjects were sacrificed at the 4 th week after the treatment beginning.The H.pylori eradication rate was judged through rapid urease test and histopathological staining,and the NF-κB expression was observed by IHC.Results: The H.pylori eradication rates was 50%(5/10) in Chenopodium ambrosioides L.group and 70%(7/10) in triple therapy group which were not statistically different(P=0.65 by Fisher probabilities).The mean optical densities of nucleus NF-κB expression and the ratios of nucleus/cytoplasm in control group were increased(P<0.05,P<0.01).After treatment,NF-κB p65 in triple therapy group and Chenopodium ambrosioides L.group was decreased in trend,but there was no significant differemce.Conclusion: Chenopodium ambrosioides L.has the bactericidal activity against H.pylori in mice,and the effects on NF-κB expression demonstrates a descending tendency while applying Chenopodium ambrosioides L..
