温肺化纤汤含药血清对小鼠肺间充质干细胞增殖和迁移能力的影响

Effects of serum containing Wenfei Huaxian Decoction on proliferation and migration of mice pulmonary mesenchymal stem cells

目的:探讨温肺化纤汤含药血清对小鼠肺间充质干细胞(PMSCs)增殖和迁移能力的影响,为温肺化纤汤临床应用治疗肺间质纤维化提供实验依据。方法:贴壁法分离培养小鼠PMSCs,流式细胞术鉴定PMSCs细胞表型并对其分化能力进行鉴定;CCK-8法检测温肺化纤汤含药血清对PMSCs增殖能力的影响;细胞划痕实验测定温肺化纤汤含药血清对小鼠PMSCs迁移能力的影响。结果:分离培养PMSCs具备间充质干细胞的细胞表型特征并具有成骨成脂分化功能。温肺化纤汤含药血清干预后,与空白对照组比较,可显著促进PMSCs的增殖和迁移能力(P<0.01,P<0.05)。结论:温肺化纤汤含药血清对小鼠PMSCs的增殖和迁移有明显的促进作用。

Objective: To investigate the effect of serum containing Wenfei Huaxian Decoction on the proliferation and migration of mice pulmonary mesenchymal stem cells(PMSCs),and to provide experimental basis for the clinical application of Wenfei Huaxian Decoction on pulmonary interstitial fibrosis.Methods: Mice PMSCs were isolated and cultured by adherence method.The phenotype and the differentiation ability of PMSCs were examined by flow cytometry.The effect of Wenfei Huaxian Decoction on the proliferation of PMSCs was examined by CCK-8 assay.The effect of Wenfei Huaxian Decoction on the migration of PMSCswas examined by cell scratch test.Results: PMSCs were isolated and cultured to express cell phenotype of mesenchymal stem cells and had osteogenic and adipogenic differentiation.Compared with the control group,serum containing Wenfei Huaxian Decoction significantly induced the proliferation and migration of PMSCs(P<0.01,P<0.05).Conclusion: Serum containing Wenfei Huaxian Decoction can promote the proliferation and migration of mice PMSCs.
