摘要
目的:进行芹菜根降血脂作用有效部位的筛选,阐明其作用机制。方法:将芹菜根提取物及分离单体加入含有AH109菌株(携带大鼠apo-AⅠ与SR-BⅠ)的培养基中,通过报告基因ɑ,β-半乳糖苷酶活力的测定来定量检测apo-AⅠ与SR-BⅠ之间互相作用力的强弱,从而反应被测药物的降脂活力大小。结果:正丁醇组分中分离出的单体能够增强两种蛋白的相互作用。结论:芹菜根中正丁醇组分中分离出的这一单体是降血脂的目标化合物。
Objective: To identify the anti-hypolipidemic effects of active component of celery root,and explore its mechanism.Methods: The extraction and the separate components of celery root were added into YPDA with AH 109 which carry apolipoprotein in AI(apo-AⅠ) and scavenger receptors class B typeⅠ(SR-BⅠ) genes of the rat.The motility of active component lipid-lowing was detected by comparing the activity of ?-galactosidase.Results: The extractions and the separate component of N-butanol extraction of celery root could obviously enhance the interaction of the apo-AⅠ protein and SR-BⅠprotein.Conclusion: The compound from n-butanol extract of celery root is a hypolipidemic target compound.
