黄芪-当归配伍对大鼠血管内膜增生模型炎性反应的影响 被引量:30

Effects of the compatibility of Astragali Radix and Angelicae Sinensis Radix on the inflammatory response of rats with intima hyperplasia

摘要 目的:研究黄芪-当归配伍对大鼠血管内膜增生炎性反应的影响。方法:将SD大鼠分为芪归1∶1组、芪归5∶1组、单用黄芪组、单用当归组、阿托伐他汀组、模型组和假手术组,采用球囊导管损伤大鼠血管内皮造成胸腹主动脉血管内膜增生模型,同时灌胃药物,连续给药14d后行腹主动脉穿刺采血分离血浆,ELISA法检测血浆炎性反应因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、单核细胞趋化因子-1(MCP-1)含量;取胸腹主动脉,免疫组化法检测血管局部细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)的表达。结果:内膜损伤14d后,血管局部和血浆中炎性反应因子含量均显著增加。与模型组比较,单用当归组、芪归1∶1组、芪归5∶1组、阿托伐他汀组血管局部ICAM-1、VCAM-1、IL-1β表达显著低于模型组(P<0.05),单用黄芪组血管局部IL-1β表达显著降低(P<0.05)。与模型组比较,阿托伐他汀组血浆中IL-1β、TNF-α、MCP-1含量显著降低(P<0.01)。结论:黄芪-当归配伍能抑制血管内膜增生时血管局部的炎性反应,其中当归是主要效应药物,黄芪-当归配伍可增强其抗血管局部炎性反应的作用。但黄芪、当归及其配伍对全身炎性反应无显著抑制作用。 Objective:To study the effect of the compatibility of Astragali Radix and Angelicae Sinensis Radix on the inflammatory response of rats with intima hyperplasia.Method:SD rats were divided into Astragali Radix Angelicae Sinensis Radix 1:1 group,Astragali Radix Angelicae Sinensis Radix 5:1 group,Astragali Radix alone group,Angelicae Sinensis Radix alone group,atorvastatin control group,model group and sham operation group.A model of intimal hyperplasia of thoracoabdominal aorta was established by balloon catheter injury in rats’vascular endothelium.Then blood was then taken to set the plasma and thoracoabdominal aorta was taken out after administration of Astragali Radix and Angelicae Sinensis Radix with different ratios for 14 days.The levels of interleukine-1β(IL-1β),tumor necrosis factor alpha(TNF-α)and monocyte chemotactic protein-1(MCP-1)in plasma were detected by ELISA.The expression of intercellular cell adhesion molecule-1(ICAM-1),vascular cell adhesion molecule-1(VCAM-1)were detected by immunohistochemistry in thoracoabdominal aorta.Results:After 14 days of intimal injury,the levels of inflammatory factors in blood vessel and plasma increased significantly.Compared with the model group,the expression of ICAM-1,VCAM-1 and IL-1βin Angelicae Sinensis Radix alone group,Astragali Radix Angelicae Sinensis Radix 1:1 group,Astragali Radix Angelicae Sinensis Radix 5:1 group,atorvastatin control group were significantly lower than those of model group(P<0.05).The expression of IL-1βin Astragali Radix alone group was significantly lower than that of the model group(P<0.05).Compared with the model group,the levels of IL-1β,TNF-αand MCP-1 in atorvastatin control group were significantly lower(P<0.01).Conclusion:Astragali Radix and Angelicae Sinensis Radix compatibility can inhibit local inflammation of blood vessel during intimal hyperplasia,of which Angelicae Sinensis Radix is the main effective drug.Astragali Radix and Angelicae Sinensis Radix compatibility can enhance its anti-inflammatory effect on local vascular inflammation.However,Astragali Radix,Angelicae Sinensis Radix and their compatibility have no significant inhibitory effect on the systemic inflammatory response.
