四君子汤对高脂饮食C57BL/6小鼠脂质代谢和组织脂蛋白酯酶基因表达的影响 被引量:13

Effects of Sijunzi Decoction on lipid metabolism and LPL gene expression in C57BL/6 mice with high fat diet

摘要 目的:观察四君子汤对高脂饮食C57BL/6小鼠脂质代谢的影响,探讨四君子汤健脾益气化痰的作用机制。方法:40只C57BL/6雄性小鼠分为正常组(8只)和高脂组(32只),分别用普通饲料和高脂饲料喂养10周。造模成功后,于第11周将高脂组随机分成模型组、四君子汤组、二甲双胍组和辛伐他汀组,每组8只。除正常组、模型组给予0.9%氯化钠溶液外,其余各组分别给予相应的药物,连续灌胃4周。每2周记录各组体质量、体长、腹围,第4、10周末眼眶静脉丛取血,第14周末摘眼球取血,进行血清总胆固醇(TG)、甘油三酯(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)检测。14周末处死小鼠,取内脏脂肪和骨骼肌检测脂蛋白酯酶(LPL)mRNA和蛋白表达。结果:干预后,各治疗组小鼠体质量、腹围及血清TG、TC、LDL-C均显著低于模型组(P<0.05,P<0.01)。小鼠内脏脂肪中,四君子汤组LPLmRNA和蛋白表达显著低于模型组(P<0.01,P<0.05)。结论:四君子汤能降低高脂饮食小鼠的体质量、腹围及血清中TG、TC、LDL-C含量,可能是通过抑制内脏脂肪中LPL表达,减少脂肪囤积,达到益气健脾化痰的目的。 Objective: To observe the effects of Sijunzi Decoction on lipid metabolism in C57 BL/6 mice fed with high fat diet and explore the mechanism of Sijunzi Decoction in invigorating spleen and benefiting qi for dissipating phlegm. Methods: Forty C57 BL/6 male mice were divided into normal(A) group(8 mice) and high-fat group(32 mice). They were fed with common and high-fat diets for 10 weeks respectively. At the 11 th week, the high-fat group was randomly divided into model group(B), Sijunzi Decoction group(C), metformin group(D) and simvastatin group(E), with 8 rats in each group. Except group A and group B were given 0.9% sodium chloride solution, the other groups were given corresponding drugs for 4 weeks. Body mass, body length and abdominal circumference were recorded every two weeks. Orbital venous plexus blood was taken at the 4 th and 10 th weekends. Eyeball blood was taken at the 14 th weekend. Serum total cholesterol(TG), triglyceride(TC), high density lipoprotein cholesterol(HDL-C), low density lipoprotein cholesterol(LDL-C) were measured. At the end of 14 weeks, mice were executed, visceral fat and skeletal muscle were taken to detect LPL gene and protein expression. Results: After intervention, the body weight, abdominal circumference and serum TG, TC, LDL-C of the mice in each treatment group were significantly lower than those in the group B(P<0.05, P<0.01). The LPL mRNA and protein expression in group C was significantly lower than that in group B in visceral adipose tissue(P<0.01, P<0.05). Conclusion: Sijunzi Decoction can reduce the body weight, abdominal circumference and serum TG, TC, LDL-C content of high-fat diet mice. Sijunzi Decoction may inhibit the expression of LPL in visceral fat and reduce fat accumulation to achieve the purpose of invigorating spleen and benefiting qi for dissipating phlegm.
