摘要
目的:探讨健脾清肠方(JPQCD)含药血清对HT29细胞内质网应激(ERS)模型NF-κB信号通路的影响健脾清肠方。方法:采用毒胡萝卜素内酯(Tg)诱导体外HT29细胞发生ERD,分为空白组(Control组),模型组(Model组)及健脾清肠方含药血清低、高浓度组(JPQCD-L组、JPQCD-H组)。用健脾清肠方含药血清干预后,Western Blot法和Real time PCR法检测HT29细胞中GRP78、NF-κB蛋白及mRNA表达。结果:与Control组比较,Model组GRP78、NF-κB蛋白及mRNA表达均增加(P<0.05);与Model组比较,JPQCD-L组和JPQCD-H组GRP78、NF-κB蛋白及mRNA表达均减少(P<0.05);与JPQCD-L组比较,JPQCD-H组GRP78、NF-κB蛋白及其mRNA表达减少(P<0.05)。结论:健脾清肠方含药血清能抑制Tg诱导体外HT29细胞发生ERS,抑制NF-κB信号通路,发挥其消炎的作用。
Objective: To investigate the effects of serum containing Jianpi Qingchang Decoction(JPQCD) on NF-κB signaling pathway in HT29 cell endoplasmic reticulum stress model. Methods: Endoplasmic reticulum stress(ERS)was induced in HT29 cells by thapsigargin(Tg), then they were divided into Control group, Model group, JPQCD drugcontaining serum low concentration group and JPQCD drug-containing serum high concentration group. After intervention with JPQCD drug-containing serum, Western Blot and Real time PCR were used to detect the protein and mRNA expressions of GRP78, NF-κB in HT29 cells. Results: Compared with the Control group, the expressions of GRP78 and NF-κB protein and mRNA in the Model group increased(P<0.05). Compared with the Model group, the expressions of GRP78 and NF-κB proteins and their mRNA in the JPQCD-L group and the JPQCD-H group were decreased(P<0.05). Compared with the JPQCD-L group, the expressions of GRP78 and NF-κB protein and mRNA in the JPQCD-H group was decreased(P<0.05).Conclusion: JPQCD-containing serum can inhibit Tg-induced ERS in HT29 cells, inhibit NF-κB signaling pathway, and exert its anti-inflammatory effect.
