温肾益气颗粒对慢性阻塞性肺疾病大鼠气道平滑肌细胞增殖及PI3K/AKT/mTOR信号通路表达的影响 被引量:17

Effects of Wenshen Yiqi Granule on the proliferation of airway smooth muscle cells and expression of PI3K/AKT/mTOR signaling pathway in chronic obstructive pulmonary disease rats

摘要 目的:研究温肾益气颗粒对香烟提取物(CSE)刺激后气道平滑肌细胞(ASMCs)增殖及PI3K/AKT/mTOR信号通路表达的影响。方法:用10%CSE刺激ASMCs。采用MTT方法检测ASMCs的增殖;采用Western blot法检测ASMCs中p-AKT、AKT、p-mTOR、mTOR以及PCNA蛋白的表达。结果:中、高剂量温肾益气含药血清能够抑制CSE刺激的ASMCs的增殖(P<0.05,P<0.01)。与模型组比较,温肾益气含药血清组ASMCs中p-AKT、p-mTOR、PCNA的蛋白表达显著降低(P<0.05)。结论:温肾益气颗粒能对COPD大鼠ASMCs的增殖起到抑制作用,其机制可能与调节PI3K/Akt/mTOR信号传导通路的表达有关。 Objective:To explore the effect of Wenshen Yiqi Granule on the proliferation of ASMCs after stimulated by cigarette smoke extract(CSE)as well as the expression of PI3 K/AKT/mTOR signaling pathway.Methods:We used 10%CSE to stimulate ASMCs.The proliferation of ASMCs were examined by MTT assay.The expression of p-AKT,AKT,p-mTOR,mTOR and PCNA proteins were detected by Western blot.Results:Wenshen Yiqi middle and high dose medicated serum can inhibit the proliferation of ASMCs with the stimulation of CSE(P<0.05,P<0.01).Compared with the model group,the expression of p-AKT,p-mTOR,and PCNA protein in ASMCs of Wenshen Yiqi medicated serum group was decreased(P<0.05).Conclusion:Wenshen Yiqi Granule can inhibit the proliferation of ASMCs in COPD rats,and its mechanism may be related to the regulation of the expression of PI3 K/Akt/mTOR signaling pathway.
