浙产铁皮石斛不同生长期石斛酚、有机酸和黄酮类成分积累变化 被引量:10

Dynamic changes of dendrophenol, organic acids and flavonoids in different growth periods of Dendrobium officinale

摘要 目的:建立铁皮石斛中石斛酚含量测定方法,并对不同生长期浙产铁皮石斛石斛酚、有机酸和黄酮类成分进行测定分析,以期从次生代谢物角度为浙产铁皮石斛采收期确定提供参考。方法:采用ACQUITY UPLCBEH C18(2.1mm×100mm,1.8μm)色谱柱,0.1%甲酸-水溶液(A)及0.1%甲酸-乙腈(B)为流动相,梯度洗脱,流速0.6mL/min,柱温40℃,正离子多反应监测(MRM)模式对石斛酚进行含量测定。结果:石斛酚在检测浓度范围内呈良好线性关系(R2=0.9997),加样回收率为95.45%,RSD值为2.1%。测定结果显示在一个生长周期内各成分总体含量表现出先降低后升高再降低的趋势,在花期及花期后达到最高;各成分分别在花期、花期后或封顶期含量最高。结论:花期及花期后(6~7月份)可作为浙产铁皮石斛潜在的最佳采收期进行更深入研究。 Objective:To establish an ultra-high-performance liquid chromatography-tandem quadrupole mass spectrometry(UPLC-MS/MS)method for the determination of dendrophenol and to determine dendrophenol,organic acids and flavonoids in different growth periods of Dendrobium officinale.Methods:The determination was carried out by an ACQUITY UPLC BEH C18(2.1 mm×100 mm,1.8μm),with 0.1%formic acid-water(A)and 0.1%formic acid-acetonitrile(B)as mobile phase for gradient elution.The flow rate was 0.6 mL/min.The column temperature was 40℃.The mass spectrometry was carried out with positive ion and multiple reaction monitoring(MRM)mode.Results:The dendrophenol in Dendrobium officinale had a good linearity(R2=0.9997)within the linear range.The recovery rate was 95.45%with RSD=2.1%.The determination results showed that the total content of ten main components decreased first then increased and decreased finally and reached the highest at about florescence.Rutin,cinnamic acid and syringate reached the highest content at top-cutting time and others components reached the highest content at about florescence.Conclusion:The peak period of secondary metabolites accumulation was from June to July,during the florescence and after the florescence period.Combined with the studies on polysaccharides,amino acids and other related primary metabolites,the study provided references for the follow-up studies on the optimal harvesting period of dendrobium Dendrobium officinale.
