补肾益髓胶囊对缓解期视神经脊髓炎谱系疾病嗜酸性粒细胞趋化因子的影响

Effects of Bushen Yisui Capsule on chemokines for eosinophil of NMOSD patients during remission

目的:观察补肾益髓胶囊对视神经脊髓炎谱系疾病(NMOSD)患者外周血中嗜酸性粒细胞CCR3受体相应趋化因子——嗜酸细胞活化趋化因子(Eot)1、Eot2、Eot3及单核细胞趋化蛋白(MCP)1、MCP2、MCP3、MCP4表达水平的影响,探讨补肾益髓胶囊通过缓解嗜酸性粒细胞活化程度治疗NMOSD的作用机制。方法:纳入缓解期NMOSD患者25例,对其进行补肾益髓胶囊干预3个月。记录研究对象的基本信息资料(年龄、首次发作年龄、病程、复发次数等),并对研究对象进行中医症状评分及扩展残疾状态量表(EDSS)评分。应用液相芯片技术检测NMOSD患者外周血中Eot1、Eot2、Eot3、MCP1、MCP2、MCP3、MCP4趋化因子的表达水平。结果:NMOSD缓解期患者经补肾益髓胶囊治疗3个月后,EDSS评分及中医症状评分显著降低(P<0.05,P<0.01);外周血Eot1、Eot2、Eot3、MCP2、MCP4较治疗前明显降低(P<0.05,P<0.01);外周血MCP1较治疗前表达未见明显差异,MCP3表达水平低于试剂盒最小测量值,未被测出。结论:补肾益髓胶囊可以明显的降低NMOSD患者的EDSS评分及中医症状评分,降低外周血嗜酸细胞活化趋化因子Eot1、Eot2、Eot3及MCP4的表达水平,同时减少MCP2趋化因子水平,减少外周血嗜酸性粒细胞、单核细胞、T细胞及DC细胞的活化程度,改善NMOSD患者的炎性环境。

Objective:To observe the effect of Bushen Yisui Capsule on eosinophil CCR3 receptor chemokineseotase-1(Eot)-1,Eot2,Eot in eosinophils in patients with Neuromyelitis optica spectrum disorders(NMOSD)3 and the effects of monocyte chemoattractant protein(MCP)1,MCP2,MCP3,MCP4 expression levels,explore the role of Bushen Yisui Capsule in the treatment of NMOSD by relieving eosinophil activation.Methods:Twenty-five patients with NMOSD during remission were enrolled,and they were treated with Bushen Yisui Capsule for 3 months.The basic information(age,first onset age,duration of disease,number of relapses,etc.)of the subjects were recorded,and the subjects were scored on the TCM symptom score and the Extended Disability Status Scale(EDSS).The expression levels of Eot1,Eot2,Eot3,MCP1,MCP2,MCP3 and MCP4 chemokines in peripheral blood of patients with NMOSD were detected by liquid-phase chip technique.Results:EDSS scores and TCM symptom scores were significantly lower in patients with NMOSD remission after 3 months of treatment with Bushen Yisui Capsule(P<0.05,P<0.01).Peripheral blood Eot1,Eot2,Eot3,MCP2,MCP4 was significantly lower than before treatment(P<0.05,P<0.01);peripheral blood MCP 1 was not significantly different from pre-treatment expression,and MCP3 expression level was lower than the minimum measurement of the kit and was not detected.Conclusion:Bushen Yisui Capsule can significantly reduce the EDSS score and TCM symptom score of NMOSD patients,reduce the expression levels of eosinophilic chemokines Eot1,Eot2,Eot3 and MCP4 in peripheral blood,and reduce the MCP2 trend.The level of factors reduces the activation of peripheral blood eosinophils,monocytes,T cells and DC cells,and improves the inflammatory environment of NMOSD patients.
