健脾清化方改善2型糖尿病大鼠肠道胰岛素抵抗

Effects of Jianpi Qinghua Formula on relieving intestinal insulin resistance in type 2 diabetes mellitus rats

目的:探讨健脾清化方对2型糖尿病(T2DM)大鼠肠道胰岛素抵抗(IR)的作用。方法:采用高脂饮食联合链脲佐菌素(STZ)建立稳定的T2DM大鼠模型,随机分为正常对照组、模型对照组和健脾清化方组,灌胃给药4周,观察各组大鼠的葡萄糖耐量和胰岛素耐量,ELISA法检测大鼠小肠组织中apoB48水平,免疫组化法检测大鼠小肠组织中GLUT2表达,Western blot法检测大鼠小肠组织中胰岛素信号通路关键因子(IRS1、IRS2、Akt)总蛋白及磷酸化水平。结果:与模型对照组比较,健脾清化方调节T2DM大鼠的葡萄糖耐量和胰岛素耐量(P<0.01),下调T2DM大鼠小肠组织中apoB48的浓度(P<0.01),增加T2DM大鼠小肠组织中GLUT2的表达,上调T2DM大鼠小肠组织中p-IRS1/总IRS1,p-IRS2/总IRS2以及p-Akt/总Akt水平(P<0.01)。结论:T2DM大鼠存在肠道IR,健脾清化方可改善T2DM大鼠的肠道IR,降低血糖。

Objective:To explore the effects of Jianpi Qinghua recipe on relieving intestinal insulin resistance(IR)in type 2 diabetes mellitus(T2 DM)Rats.Methods:The rat model of T2 DM was established by a combination of small dosage of streptozotocin(STZ)injection with high-fat diet.Rats were randomly divided into the control group,model group and Jianpi Qinghua Formula group.All rats were treated by gavage for 4 weeks.Intraperitoneal glucose tolerance test(IPGTT)and intraperitoneal insulin tolerance test(IPITT)were carried out.The concentration of apoB48 from intestinal tissues was detected by ELISA.The expression of GLUT2 from intestinal tissues were determined by immunohistochemistry.The expression of IRS1,IRS2 and Akt,which play vital roles in insulin signaling pathway in rats’intestinal tissues,were tested by Western blot.Results:Compared with the model group,Jiangpi Qinghua Formula regulated glucose tolerance and insulin tolerance in T2 DM rats,decreasing the level of apoB48(P<0.01)and increasing the expression of GLUT2 in the intestinal tissue of T2 DM rats.Jianpi Qinghua Formula also up-regulated the level of p-IRS1/total IRS1,p-IRS2/total IRS2 and p-Akt/total Akt in the intestinal tissue from T2 DM rats(P<0.01).Conclusion:T2 DM rats have intestinal IR.Jiangpi Qinghua Formula can reduce blood glucose by relieving intestinal IR in T2 DM Rats.
