摘要
目的:观察加味逍遥丸对更年期大鼠行为学和神经-内分泌的影响,探讨其治疗更年期综合征的作用机制。方法:90只雌性SD大鼠随机分为假手术组、模型组、戊酸雌二醇组和加味逍遥丸高、中、低剂量组。除假手术组外,其余各组大鼠摘除双侧卵巢复制更年期动物模型,给药后分别进行动物行为学测评,同时测定大鼠血清相关性激素水平和下丘脑中神经递质的含量。结果:与模型组相比,加味逍遥丸高、中、低剂量组均可显著增加更年期大鼠在十字迷宫中的相对开臂次数(P<0.05,P<0.01);加味逍遥丸高、中剂量组可显著增加更年期大鼠的明箱相对活动时间和穿格次数(P<0.05,P<0.01);加味逍遥丸高、中剂量组可显著降低更年期大鼠下丘脑中5-HT的含量(P<0.05);加味逍遥丸高、中、低剂量组可显著升高更年期大鼠下丘脑中NA和DA含量(P<0.01,P<0.05)。结论:加味逍遥丸对更年期模型动物的行为学改变具有较好的改善作用,具有较好的改善和调节模型动物中枢单胺类神经递质紊乱状态的作用。
Objective:To observe the effects of Jiawei Xiaoyao(JW)Pills on ethology and nerve-endocrine in climacteric rats,and to discuss its regulation mechanism of JW Pills on the climacteric syndrome.Methods:A total of 90 sexually mature SD female rats were randomly divided into sham-operated group,climacteric model group,estradiol valerate group,JW Pills high dose,medium dose and low dose groups.In addition to the sham-operated group,other groups were removed the bilateral ovarian to replicate climacteric animal model.After the administration,the rats were subjected to ethology assessment.Related hormone levels in serum and neurotransmitters in the hypothalamus were determined.Results:Compared with climacteric model group,JW Pills high dose,medium dose and low dose groups could obviously increase the climacteric rats’index of open arm entry in elevated plus-maze test(P<0.05,P<0.01),JW Pills high dose and medium dose groups could obviously increase the climacteric rats’relative activity time of lucida and frequency of pass in light-dark box(P<0.05,P<0.01).JW Pills high dose and medium dose groups could decrease elevated content of 5-HT in hypothalamus due to ovariectomy(P<0.05).JW Pills high dose,medium dose and low dose groups showed significant difference in increasing the NA and DA contents in ovariectomized rats(P<0.01,P<0.05).Conclusion:JW Pills can regulate ethology of climacteric rats,and has a better effect on improving and regulating the disorder of monoamine neurotransmitters in the model animals.
