金银花和黄芩不同煎煮方式化学成分变化及抗病毒作用比较

Comparison of ingredients and anti viral effects of different decoctions methods of Lonicera japonica Thunb. and Scutellaria baicalensis Georgi

目的:分析银黄制剂不同煎煮方式化学成分的变化,比较金银花和黄芩分煎与合煎体外抗病毒作用差异。方法:采用高效液相色谱法建立分析金银花和黄芩合煎与分煎化学成分变化的方法。以人喉癌上皮细胞(Hep2)作为宿主细胞,并用呼吸道合胞病毒(RSV),柯萨奇病毒B3、B5(COXB3、COXB5),肠道病毒71型(EV71)4种病毒感染。用金银花和黄芩的分煎液与合煎液作为4种病毒感染细胞的治疗药物,阳性对照药采用利巴韦林。根据细胞病变效应法(CPEF法)结合CCK-8染色法检测细胞存活率,计算药物的治疗指数TI值。结果:金银花和黄芩不同煎煮方式对化学成分产生了较大变化,金银花和黄芩的分煎液与合煎液均有不同程度的抗RSV、COXB3、COXB5、EV71病毒活性;其中对RSV的抑制作用最好,金银花和黄芩的分煎液对RSV、COXB3、COXB5、EV71的治疗指数均大于其合煎液。结论:金银花和黄芩不同煎煮方式对其中成分产生的影响较大,金银花和黄芩的分煎液对4种病毒的抑制作用均优于合煎液,其中对RSV的抑制作用最好。

Objective:To analyze the changes of chemical components and compare the anti viral effects of different decocting methods of Lonicera japonica Thunb.and Scutellaria baicalensis Georgi in vitro.Methods:High performance liquid chromatography(HPLC)was used to establish a method for the analysis of chemical composition in the decoction of honeysuckle and Scutellaria baicalensis.Human laryngeal cancer epithelial cells(Hep2)were used as host cells and infected with respiratory syncytial virus(RSV),coxsackie virus B3,B5(COXB3,COXB5)and entero virus 71(EV71).The decoctions of Lonicera japonica Thunb.and Scutellaria baicalensis Georgi were used as therapeutic drugs for four kinds of virus-infected cells,and ribavir in was used as positive control drug.Cell viability was measured by cytopathic effect(CPE)and CCK-8 staining,and the therapeutic index TI was calculated.Results:Different decoction methods of Lonicera japonica Thunb.and Scutellaria baicalensis Georgi had great changes in chemical composition.The decoction of Lonicera japonica Thunb.and Scutellaria baicalensis Georgi had different degrees of anti-RSV,COXB3,COXB5 and EV71 activity,among which RSV inhibition was the best.The decoction of Lonicera japonica Thunb.and Scutellaria baicalensis Georgi had the best effect on RSV,COXB3,COXB5 and EV71.The number of decoctions was greater than that of the combined decoction.Conclusion:The different decocting methods of Lonicera japonica Thunb.and Scutellaria baicalensis Georgi have great influence on their components.The inhibition effect of the four viruses is better than that of the combined decoction,and RSV is the best.
