响应面法协同正交试验设计优化薏苡仁多糖提取工艺研究 被引量:16

Optimization of extraction process conditions of coix seed polysaccharide by response surface method and orthogonal design of experiment

摘要 目的:确定薏苡仁多糖最佳提取工艺。方法:在单因素试验和正交设计的基础上,应用Box-Behnken中心组合试验设计和响应面(RSM)分析法,以薏苡仁多糖得率为响应值绘制响应面图和等高线图,回归分析薏苡仁多糖提取的影响因素,优选薏苡仁多糖提取工艺。结果:正交设计与响应面法优化结果接近,所得薏苡仁多糖的最佳提取条件为:水浸提时间75min,水提取温度92℃,料液比(g/L)5∶1。结论:此方法对薏苡仁多糖提取条件的优化合理可行,为简化薏苡仁提取工艺以及提高薏苡仁多糖得率提供了依据,且预测性良好。 Objective:To determine the optimum extraction process of coix seed polysaccharide.Methods:On the basis of single factor test and orthogonal design,Box-Behnken center combination test design and response surface(RSM)analysis method were used to draw response surface maps and contour plots with the yield of coix seed polysaccharide as response value.The influencing factors of coix seed polysaccharide extraction were analyzed by regression analysis,and the extraction process of coix seed polysaccharide was optimized.Results:The results of orthogonal design and response surface method were close to each other.The optimal extraction conditions of coix seed polysaccharide were as follows:water extraction time 75 min,water extraction temperature 92°C,and solid-liquid ratio(g/L)5:1.Conclusion:This method is reasonable and feasible for the optimization of the extraction conditions of coix seed polysaccharide,which provides a basis for simplifying the extraction process of coix seed and improving the yield of coix seed polysaccharide,and it has good predictability.
