当归四逆汤对血管平滑肌细胞中ERK1、c-myc蛋白表达的影响 被引量:2

Effects of Danggui Sini Decoction on ERK1, c-myc protein expression in vascular smooth muscle cells

摘要 目的:观察当归四逆汤对血管平滑肌细胞中ERK1、c-myc蛋白表达的影响,探讨其防治动脉硬化闭塞症(ASO)的作用机制。方法:以体外培养兔主动脉血管平滑肌细胞(VSMC)为研究对象,应用血清药理学法,以血管紧张素Ⅱ(AngⅡ)为诱导因素,用当归四逆汤进行干预,采用Western Blot法检测兔血管平滑肌细胞中ERK1、c-myc蛋白的表达。结果:与正常组比较,模型组ERK1、c-myc蛋白表达水平显著升高;与模型组比较,各用药组ERK1、c-myc蛋白表达水平显著降低,其中高剂量组ERK1、c-myc蛋白表达水平降低最为明显,低剂量组降低最不明显。结论:当归四逆汤通过抑制原位癌基因c-myc的表达和细胞信号通路ERK1的表达,抑制了血管平滑肌细胞的增殖和迁移,阻止了ASO的发生和发展。 Objective:To discuss the mechanism of Danggui Sini Decoction control atherosclerosis obliterans(ASO)by observing the effect of Danggui Sini Decoction on ERK1,c-myc protein expression in vascular smooth muscle cells.Methods:In vitro culture of rabbit aortic VSMC as the research object,the application of serum pharmacological method,setting angiotensin II(Ang II)for inducing factors,four decoction intervene with angelica,using Western Blot method to detect the rabbit vascular smooth muscle cells in the expression of ERK1,c-myc protein.Results:Compared with normal serum group,model group ERK1,c-myc protein expression level increased significantly;Compared with model group,the treatment group ERK1,c-myc protein expression level decreased obviously,which Danggui Sini Decoction high dose group of ERK1,c-myc protein expression levels is most obvious,the reduce in low dose group is not obvious.Conclusion:Danggui Sini Decoction control ASO by inhibiting the expression of ERK1,c-myc protein and then inhibiting vsmc proliferation and migration.
