Study on the antiviral effective components of Euonymus Alatus in vitro

摘要 目的:探索鬼箭羽不同提取方法对不同病毒的体外抑制作用,并确定最佳分离纯化效果的大孔树脂型号。方法:分别用蒸馏水、95%乙醇、乙酸乙酯提取鬼箭羽,收集浓缩液。采用细胞体外病毒感染模型,进行不同病毒体外抗病毒作用评价,通过显微镜下观察细胞病变效应,判断不同提取方法对不同病毒的抑制作用;通过大孔树脂吸附法进行纯化分离,确定最佳大孔树脂型号。结果:经过初步筛选鬼箭羽水提醇沉后的上清液对RSV的抑制作用最强,TI值为34.91;HPD-100树脂对鬼箭羽的分离纯化效果最佳,水洗脱部位TI值为52.000。结论:确定其最敏感菌株为呼吸道合胞病毒,并确定其水提醇沉上清液部分的效果最佳,经HPD-100吸附后水洗脱液的抗病毒效果有了明显提高。 Objective:To explore the inhibitory effects of different extraction methods on different viruses in vitro,and to determine the best macroporous resin type for separation and purification effect.Methods:Extract Euonymus Alatus with distilled water,95%ethanol and ethyl acetate,and collect concentrated solution.The virus infection model in vitro was used to evaluate the antiviral effect of different viruses in vitro.The cytopathic effect(CPE)was observed under the microscope to determine the inhibitory effect of different extraction methods on different viruses.After purification and separation by macroporous resin adsorption method,the best macroporous resin type was determined.Results:After preliminary screening,the supernatant after water extraction and ethanol precipitation had the strongest inhibitory effect on RSV,with TI value of 34.91.HPD-100 resin had the best separation and purification effect,with TI value of 52.000 at the water elution site.Conclusion:The most sensitive strain is identified as respiratory syncytial virus,and the best effect was obtained by water extraction and ethanol precipitation of supernatant.After HPD-100 adsorption,the antiviral effect of water eluate has been significantly improved.
