腺苷A1受体介导异丙酚预处理对局灶性脑缺血再灌注损伤大鼠的缺血耐受 被引量:2

摘要 目的:观察腺苷A1受体是否介导异丙酚预处理对局灶性脑缺血再灌注损伤大鼠的缺血耐受。方法:SD大鼠随机分为3组:对照组、异丙酚组、溶剂组(n=6),采用高效液相色谱法(HPLC)检测各组脑皮质区腺苷浓度。另一批SD大鼠随机分成A:模型组(MCAO模型),B:腺苷A1受体拮抗剂(DPCPX)组,C:异丙酚组,D:溶剂组(脂肪乳剂组),E:异丙酚+DPCPX组,F:异丙酚+拮抗剂溶剂组(DMSO)(n=6)。结果:与对照组比较,异丙酚组脑皮质区腺苷浓度明显升高(P<0.05)。与C组比较,A、D、E组大鼠行为学评分和脑梗死体积均明显增加(P<0.05,P<0.05,P<0.05),B、F组比较无差异。结论:异丙酚预处理对脑缺血再灌注损伤的缺血耐受作用可能部分由腺苷A1受体介导。 AIM: To investigate whether adenosine A1 receptor mediate neuroprotection induced by propofol pretreament after focal cerebral ischemia reperfusion injury in rats. METHODS: To determine the effect of propofol pretreatment on the adenosine concentration in rat. Eighteen male SpragueDawley( SD) rats were randomly divided into 3groups( n = 6) : control group,propofol group and vehicle group. The adenosine concentration was measured by high performance liquid chromatography( HPLC). Thirty-six male SD rats were randomly allocated into six groups( n = 6) : MCAO group;DPCPX group; propofol group; Intralipid group;propofol + DPCPX group and propofol + DMSO group. RESULTS: Compared with the control group,propofol cerebral cortex adenosine concentrations were significantly increased( P 〈0. 05). Compared with propofol group, the behavior of rats scores and infarct volume of MCAO,intralipid and propofol + DPCPX group were significantly increased( P 〈0. 05,P 〈0. 05,P 〈0. 05),no difference in DPCPX and propofol + DMSO group. CONCLUSION: Ischemia tolerance induced by propofol pretreatment to focal cerebral ischemia reperfusion may be partially mediated by the adenosine A1 receptor.
