血浆游离DNA中TGR5、RIZ1异常甲基化在肝癌诊断中的价值

摘要 目的研究外周血浆游离DNA中TGR5、RIZ1异常甲基化在肝细胞癌(HCC)诊断中的价值。方法收集48例HCC和40例健康体检者的血浆,提取血浆游离DNA,运用甲基化特异性PCR(MSP)检测血浆游离DNA中TGR5、RIZ1甲基化状态,结合其病理资料进行分析。结果HCC组血浆游离DNATGR5、RIZ1甲基化率分别为68.8%和60.4%;两者联合检测时HCC组甲基化率为72.9%。正常对照组甲基化率分另IJ5.0%和2.5%。TGR5、RIZ1甲基化状态与HCC患者的性别、年龄、肿瘤大小、肿瘤数目、HBeAg阳性及TNM分期等病理参数无关(P〉0.05)。结论TGR5、RIZ1甲基化水平在早期HCC血浆中升高,通过联合检测患者TGR5、RIZ1甲基化可作为HCC的诊断指标。 Objective To study the value of abnormal methylation of TGR5 and RIZ1 of free DNA in peripheral plasma in diagnosis of hepatocellular carcinoma (HCC). Methods The plasma samples of 48 patients with HCC and 40 normal controls were collected and free DNA was extracted from the plasma. The methylation statuses of TGR5 and RIZ1 were determined using methylation specific PCR method. Results The methylation rates of TGR5 and RIZ 1 in the HCC group were 68.8% and 60.4%, respectively, which were significantly higher than those (5.0% and 2.5%) in the control group (P〉0.05). The methylation rate of the combination of TGR5 and RIZI in the HCC group was 72.9%. The methylation status of TGR5 and RIZ1 didn't correlated with sex, age, tumor diameter, lymph node metastases, and TNM stage(/〉〉0.05). Conclusions The methylation levels of TGR5 and RIZ1 increase in plasma in early stage of HCC. The detection of the methylation of both TGR5 and RIZ1 is a choice for the early diagnosis of HCC.
