摘要 目的对杜仲Eucommia ulmoides进行血清药物化学研究。方法采用UHPLC-Q-TOF-MS技术,通过比较杜仲提取物、给药组及空白组大鼠血清的指纹图谱,确定口服杜仲提取物后大鼠血中移行成分。结果给药组大鼠血清指纹图谱与空白组血清指纹图谱对比有明显不同。含药血清中出现7个移行成分,与杜仲提取物指纹图谱在相同保留时间出峰,为原型成分。采用对照品对照,确定5个色谱峰所表征的化学成分为原型吸收入血成分,依次为京尼平苷酸、原儿茶酸、京尼平苷、松脂醇二葡萄糖苷、松脂醇单葡萄糖苷。通过查阅文献,推断了2个色谱峰所表征的化学成分可能为1-羟基松脂醇二葡萄糖苷、杜仲醇。结论血中移行成分可能为杜仲的体内直接作用物质,为明确杜仲药效物质基础提供了科学依据。 Objective To investigate the bioactive constituents in the stem bark of Eucommia ulmoides and study on their serum pharmacchemistry. Methods The UHPLC-Q-TOF-MS was used to analyze the ingredients in the serum samples in rats, and the chromatogram was compared among the peaks of extracts in the stem bark of E. ulmoides, rat serum with drug and blank serum sample was used to determine them after the administration of extracts by comparing the finger-print. Results Seven compounds absorbed into blood were be detected, and had the same retention time with those in the finger-print of extracts, after repeated experiments, which were original constituents of the extracts. The extracts were identified by comparing the retention times with standard substance, five compounds were obtained and identified as geniposidic acid, protocatechuic acid, geniposide, pinoresinol diglucoside, and pinoresinol monoglucoside. Through review of the literature, two compounds may be 1-hydroxypinoresinol glucoside, and eucommiol. Conclusion The compounds are absorbed into the blood are the effective constituents, and the research provides a scientific fundament for material basis of effectiveness of E. ulmoides.
