俄色叶-A毛囊靶向脂质体制备工艺的优化 被引量:2

摘要 目的优化俄色叶-A毛囊靶向脂质体的制备工艺。方法薄膜-超声法制备脂质体。以包封率为指标,单因素试验和Box-Behnken效应面法优化制备工艺。结果最佳参数为卵磷脂与胆固醇比例5∶1,卵磷脂与药物比例10∶1,缓冲液用量15 m L。包封率的预测值和实测值分别为91.8%和92.3%。脂质体粒子呈球形,粒径为(162.1±6.3)nm,Zeta电位为(-32.8±7.3)m V。结论该制备工艺简单可行,包封率高。 AIM To optimize the preparation process of eseye-A hair follicle-targeted liposomes. METHODS The liposomes were prepared by film-ultrasonic method. With encapsulation efficiency as an index,the preparation process was optimized by single factor experiment and Box-Behnken response surface method.RESULTS The optimized parameters were 5 ∶ 1 for ratio of lecithin to cholesterol,10 ∶ 1 for ratio of lecithin to drug and 15 m L for the amount of buffer solution. The predictive value and measured value were 91. 8 % and92. 3 %,respectively. The spherical liposome particle had particle size of( 162. 1 ± 6. 3) nm and Zeta potential of(-32. 8 ± 7. 3) m V. CONCLUSION This simple and feasible preparation process ensures a high encapsulation efficiency.
