摘要 目的建立HPLC法测定疱疹康喷雾剂(大黄、黄连、白芷等)中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚、盐酸小檗碱的含有量。方法盐酸小檗碱的分析采用Venusil XBP C18(L)色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈-水-冰乙酸-三乙胺(28∶70∶1∶1);检测波长265 nm;柱温25℃;体积流量1 m L/min。其他5种成分的分析采用Venusil XBP C18(L)色谱柱(4.6 mm×250 mm,5μm);流动相为甲醇-0.1%磷酸溶液(75∶25);检测波长254 nm;柱温25℃;体积流量1 m L/min。结果芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚、盐酸小檗碱分别在0.017 9~0.358μg(r=0.999 9)、0.018 2~0.364μg(r=0.999 8)、0.018 9~0.338μg(r=0.999 9)、0.008 0~0.160μg(r=1)、0.008 9~0.177μg(r=0.999 9)、0.147~1.470μg(r=0.999 7)范围内线性关系良好,平均回收率分别为101.3%(RSD=1.0%)、102.8%(RSD=1.5%)、101.2%(RSD=1.9%)、102.7%(RSD=1.3%)、100.2%(RSD=1.9%)、101.3%(RSD=2.0%)。结论该方法灵敏、简便、准确、重复性好,可用于疱疹康喷雾剂的质量控制。 AIM To establish an HPLC method for determining the contents of aloe-emodin,rhein,emodin,chrysophanol,physcion and berberine hydrochloride in Paozhenkang Spray( Rheum palmatum,Coptis chinensis,Angelica dahurica,etc.). METHODS The analysis of berberine hydrochloride was conducted on Venusil XBP C18( L)column( 4. 6 mm × 250 mm,5 μm) with its temperature maintained at 25 ℃,a mobile phase of acetonitrile-water-acetic acid-triethylamine( 28 ∶ 70 ∶ 1 ∶ 1),265 nm for detection wavelength,and flow rate of1 m L / min. The analyses of another five constituents were performed on Venusil XBP C18( L)column( 4. 6 mm ×250 mm,5 μm) and a mobile phase comprising of methanol-0. 1% phosphoric acid solution in ratio 75 ∶ 25.The other conditions were 254 nm detection for wavelength,25 ℃ for column temperature,and 1 m L / min for flow rate. RESULTS Aloe-emodin,rhein,emodin,chrysophanol,physcion and berberine hydrochloride showed good linear relationships within the ranges of 0. 017 9-0. 358 μg( r = 0. 999 9),0. 018 2-0. 364 μg( r = 0. 999 8),0. 018 9-0. 338 μg( r = 0. 999 9),0. 008 0-0. 160 μg( r = 1),0. 008 9-0. 177 μg( r = 0. 999 9) and0. 147-1. 470 μg( r = 0. 999 7),whose average recoveries were 101. 3%,102. 8%,101. 2%,102. 7%,100. 2% and 101. 3% with the RSDs of 1. 0%,1. 5%,1. 9%,1. 3%,1. 9% and 2. 0%, respectively.CONCLUSION This sensitive,accurate,simple and reproducible method qualifies in the quality control of Paozhenkang Spray.
