摘要 目的:探讨丹参多糖(SMPS)对急性肝损伤小鼠肝窦内皮细胞(LSEC)窗孔的影响。方法:尾静脉注射脂多糖(LPS)诱导小鼠急性肝损伤模型,检测血清中谷草转氨酶(AST)、谷丙转氨酶(ALT)、一氧化氮(NO)的水平,比较肝组织中小窝蛋白-1(Car-1)的表达情况及LSEC窗孔的改变情况。结果:SMPS能降低血清中AST、ALT、NO的含量,减少Car-1的表达,增加LSEC的数目,并其使窗孔孔径增大。结论:SMPS通过开放LSEC窗孔,改善肝窦微循环,达到保护肝脏的作用。 Objective:To investigate the effect of Salvia miltiorrhiza polysaccharides(SMPS) on sinusoidal endothelial cell(LSEC) fenestrae in mice with acute liver injury.Methods:Mice models with acute liver injury were induced by lipopolysaccharide(LPS) through tail vein injection.The levels of serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),and nitric oxide(NO) were determined.The changes of caveolin-1(Cav-1) expression and LSEC fenestrae in liver tissues were compared.Results:SMPS could decrease the content of serum AST,ALT and NO,reduce the expression of Cav-1,and increase the number of LSEC and the diameters of fenestrae.Conclusion:SMPS can improve sinusoidal microcirculation by opening LSEC fenestrae to protect liver.
