摘要
为提高罗非鱼加工下脚料的综合利用率,开发新型的食品级蛋白酶制剂,对罗非鱼肠道蛋白酶的提取及催化特性进行了探讨.试验结果表明,罗非鱼肠道中含有非常丰富的碱性蛋白酶,其提取率可达到6200U/g;该蛋白酶在pH9.0、温度60℃的条件下有最大催化活性;在pH7~9的范围内,温度低于50℃的条件下有很好的稳定性;Fe2+离子、PMSF及EDTA对该酶表现出较强的抑制作用,初步显示该酶为金属离子依赖型丝氨酸蛋白酶.
To improve the comprehensive utilization of the Tilapia viscera and develop a new type of food grade protease, the extraction and catalytic properties of protease from Tilapia intestine were investi- gated. The results showed that the content of protease in the tilapia intestine was very rich, and the ex- traction yield of protease from tilapia intestine could reach 6000U/g; the protease extracted from tilapia in- testine was one alkaline protease which had maximum activity at pH 9.0 and 60 ℃ ;the protease had good stability at pH 7~9 and temperature ~ 50℃ ; Fe^2+ and EDTA could partially inhibit the activity of the protease, which indicated this enzyme was a metal ion dependent serine protease.
