摘要 目的研究高良姜不同活性部位对兔离体肠管运动的影响和作用机制。方法采用减压硅胶柱层析法,对高良姜乙醇提取物进行分离;借助薄层色谱法及高效液相指纹图谱,对各活性部位进行归属与鉴别;观察高良姜活性组分对离体兔正常肠肌和乙酰胆碱诱导肠管痉挛的作用。结果高良姜提取物中活性组分均可剂量依赖性地抑制肠肌自发性运动,其中黄酮类、二苯基庚烷类最明显(P<0.05),比山莨菪碱作用更强(P<0.05),各活性组分能抑制乙酰胆碱诱导肠管痉挛。结论高良姜各活性组分均可直接抑制离体肠肌张力和非竞争性拮抗乙酰胆碱的作用,其中黄酮类效果最强。 Objective To investigate the effect of active components of galangal extract on smooth muscle of isolated intestine and possible mechanisms in rabbits. Methods The ethanol extract of galangal was separated by vacuum liquid chromatography,and identified by thin layer chromatography and high performance liquid chromatography. The role of active components of galangal extract in normal movement and acetylcholine-induced contraction of smooth muscle was observed in the isolated rabbit intestine. Results The active components of galangal extract inhibited the spontaneous movement of intestinal smooth muscle in a dose-dependent manner,of which flavonoids and diphenylheptanes showed more effective than anisodamine(P〈0.05). Meanwhile,they suppressed acetylcholine-induced contraction of intestine. Conclusion The active components of galangal extract can directly inhibit the muscular tension of the isolated intestine and noncompetitively antagonize the role of acetylcholine,with flavonoids being the most effective.
