肿瘤增殖相关分子(TSPAN-1)和血管内皮生长因子C(VEGF-C)在宫颈鳞癌中的表达及意义

目的通过检测肿瘤增殖相关分子(TSPAN-1)和血管内皮生长因子C(VEGF-C)在正常宫颈(NCE)组织、宫颈上皮内瘤变(CIN)及宫颈鳞癌(SCC)组织中的表达情况,探讨两者在宫颈鳞癌发生、发展中的作用及意义。方法采用免疫组织化学SP法检测15例NCE组织,30例高级别CIN组织及45例SCC组织中TSPAN-1和VEGF-C的表达情况,分析两者的表达与宫颈癌患者各临床因素的关系。结果 TSPAN-1和VEGF-C在正常宫颈组织中均不表达;在CINI I+CINI I级、宫颈鳞状细胞癌组织中两者的表达明显升高。TSPAN-1在宫颈鳞癌的阳性表达率与患者年龄、FIGO分期、组织分化程度无关,与淋巴结转移和宫颈浸润深度有关(P<0.05)。VEGF-C在宫颈鳞癌组中的阳性表达率与患者年龄、FIGO分期无关,而与宫颈浸润深度、组织分化程度、淋巴结转移有关(P<0.05)。宫颈鳞癌组织中TSPAN-1和VEGF-C的表达呈正相关(r=0.851,P<0.05)。结论 TSPAN-1和VEGF-C在正常宫颈组织的表达均明显低于宫颈上皮内瘤变组、宫颈鳞癌组中的表达。在宫颈鳞癌组中TSPAN-1的阳性表达率与年龄、FIGO分期、组织分化程度无关,而与间质浸润深度及淋巴结转移相关。VEGF-C在宫颈鳞癌组中的阳性表达率与年龄、FIGO分期无关,而与浸润深度、组织分化程度、淋巴结转移密切相关。TSPAN-1及VEGF-C在宫颈鳞癌中的表达呈正相关,说明两者可能与宫颈鳞癌的发生、发展相关。

Objective To study the expression oftetraspanin-1 and vascularendothelial growth factor-C in the normal cervical epithelium （NCE）, cervical intraepithelial neoplasia （C1N） and squamous carcinomas of cervix （SCC）, and to investigate the relationship of TSPAN-1 and VEGF-C in the occurrence and development of squamous carcinomas of cervix. Methods Immunohistochemistry SP was employed to detect the expression of TSPAN-1 and VEGF-C in 45 cases of SCC, 30 cases of high level CIN and 15 cases of NCE. This study was to explore the relationships between the two kinds of protein expression and the clinical pathological parameters of SCC. Results In the process of normal cervix to the cervical intra- epithelial neoplasia to cervical squamous cancer, the expression of TSPAN-1 and VEGF-C increased gradually. The positive expression rates of TSPAN-1 were not associated with clinical pathological index of SCC such as age, FIGO stages, historical differentiation, but associated with the depth of tumor invasion and lymph node metastasis. The positive expression rates of VEGF-C in SCC were not associated with clinical pathological index of SCC such as age, FIGO stages, but associated with historical differentiation, the depth of tumor invasion and lymph node metastasis. The expression level of TSPAN-1 and VEGF-C in cervical squamous cancer were positively correlated. Conclusion The expression of TSPAN-1 and VEGF-C in the cervical tissue of different research object; in the process of normal cervix to the cervical intraepithelial neoplasia to cervical squamous cancer the expression of TSPAN-1 and VEGF-C increased gradually, but the positive expression level of TSPAN-1 and VEGF-C in cervical squamous Cancer have non-significant higher than it in the cervical intra-epithelial neoplasia. There was no correlation between the expressions of TSPAN-1, VEGF-C and the pathoclinical parameters, including the age, FIGO stages, but associated with the depth of tumor invasion and lymph node metastasis in SCC. The expression of TSPAN-1 was correlated with that of VEGF-C in SCC.
