摘要 目的探讨IL-23对小鼠B16黑色素瘤细胞VEGF表达水平的影响。方法体外培养小鼠B16黑色素瘤细胞,选择对数生长期的细胞,在培养基中加入不同浓度(5 ng/ml、10 ng/ml、20 ng/ml、50 ng/ml)的IL-23,并设空白对照组,48 h后通过ELISA实验检测IL-23对B16黑色素瘤细胞VEGF表达水平的影响。结果不同浓度IL-23处理组与对照组相比,B16黑色素瘤细胞培养上清液中VEGF的表达水平明显升高,且随着IL-23浓度的增加,VEGF的含量也增加(P<0.05),呈浓度依赖性。结论 IL-23可促进小鼠B16黑色素瘤细胞分泌VEGF。 Objective To investigate the influence of IL-23 on the levels of VEGF expressed by mouse B16 melanoma cells. Methods Mouse B16 melanoma cells were cultured in vitro. Cells in research group were treated with various concentrations(5 ng/ml, 10 ng/ml, 20 ng/ml, 50 ng/ml) of recombinant mouse IL-23 and those in control group without IL-23.The levels of VEGF expressed by B16 melanoma cells were detected by ELISA after 48 hours. Results The VEGF levels in the supernatants of the B16 cells treated with various concentrations of IL-23 were higher than that in the control group by ELISA(P〈0.05), which was IL-23 concentration-dependent. Conclusion IL-23 can promote the mouse B16 melanoma cells secreting VEGF.
